The study of isolation, characterization, and bioactivity tests of alkaloid compound from sponge have been carried out. Bioactivity test was examined by using chloramphenicol-resistant Staphylococcus aureus. The result of screening toward five types of sponges showed that sponge of Callyspongia sp. has the greatest antibacterial activity. The bioactive metabolite was isolated through several steps chromatography and was purified by Medium Pressure Liquid Chromatography (MPLC) acquired 2 mg (0.001%) of bioactive substance M4T. M4T revealed as an amorphous crystal. Identification using Thin Layer Chromatography (TLC) with Dragendorff’s reagent showed orange spot which indicated as alkaloid compound. The characterization of M4T using UV-Vis spectrophotometer exhibited maximum absorption at a wavelength of 202 nm (transition of n → π *) and 279 nm (transition of π → π *). Interpretation of IR spectrum indicated the presence of hydroxyl functional groups O-H (3441 cm-1, 1057 cm-1), alkane C-H (2924 cm-1, 2854 cm-1), carbonyl C=O (1720 cm-1), alkene C=C (1651 cm-1, 1558 cm-1), and tertiary amines C-N (1381 cm-1, 1249 cm-1). The result of bioactivity test at dose 50 μg showed inhibition zone with a diameter of 14 mm against Staphylococcus aureus which resistant to chloramphenicol. Based on the results, M4T is an alkaloid compound which potential as antibacterial. Keywords: Alkaloids, sponge, antibacterial, Staphylococcus aureus. Telah dilakukan studi isolasi, karakterisasi, dan uji bioaktivitas senyawa alkaloid dari spons. Uji bioaktivitas dilakukan terhadap bakteri Staphylococcus aureus yang resisten terhadap kloramfenikol. Hasil skrining terhadap lima jenis spons diperoleh bahwa spons Callyspongia sp. memiliki aktivitas antibakteri paling besar. Proses isolasi bioaktif metabolit dilakukan melalui beberapa tahapan kromatografi dan pemurnian menggunakan Medium Pressure Liquid Chromatography (MPLC) diperoleh zat aktif M4T sebanyak 2 mg (0,001%). Senyawa M4T berbentuk kristal amorf. Identifikasi menggunakan Kromatografi Lapis Tipis (KLT) dengan pereaksi Dragendorff, menunjukkan bercak warna jingga yang mengindikasikan golongan alkaloid. Karakterisasi M4T mengggunakan spektrofometer UV-Vis menunjukkan adanya serapan pada panjang gelombang maksimum 202 nm (transisi n → π*) dan 279 nm (transisi π → π*). Hasil interpretasi spektrum IR menunjukkan adanya gugus fungsi hidroksi O-H (3441 cm-1, 1057 cm-1), alkana C-H (2924 cm-1, 2854 cm-1), karbonil C=O (1720 cm-1), alkena C=C (1651 cm-1, 1558 cm-1), dan amina tersier C-N (1381 cm-1, 1249 cm-1). Hasil uji bioaktivitas M4T memberikan zona hambat dengan diameter 14 mm terhadap bakteri Staphylococcus aureus resisten terhadap kloramfenikol pada dosis 50 μg. Berdasarkan data yang diperoleh, M4T merupakan senyawa alkaloid yang memiliki potensi sebagai antibakteri. Kata Kunci: Alkaloid, spons, antibakteri, Staphylococcus aureus,
