Abstrak (Berisi Bastraknya saja, Judul dan Nama Tidak Boleh di Masukan)

Abstrak Pembesaran seedling anggrek Dendrobium in vitro dapat menggunakan dua formulasi media dasar yaitu media ½ MS dan media Growmore. Penelitian ini bertujuan untuk mengetahui pengaruh (1) media dasar ½ MS atau media Growmore yang menghasilkan pertumbuhan seedling anggrek Dendrobium yang lebih baik, (2) bahan adenda yang menghasilkan pertumbuhan seedling anggrek Dendrobium paling baik, (3) kombinasi media dasar dan bahan adenda mana yang menghasilkan pertumbuhan seedling anggrek Dendrobium terbaik. Penelitian dilaksanakan di Laboratorium Kultur Jaringan Tanaman, Fakultas Pertanian, Universitas Lampung dimulai bulan November 2010 sampai bulan Mei 2011. Penelitian ini menggunakan rancangan teracak sempurna (rts) dengan rancangan perlakuan faktorial (2x4). Faktor pertama adalah media ½ MS atau media Growmore dan faktor kedua adalah jenis bahan adenda yaitu ekstrak tomat, ekstrak kentang, ekstrak taoge, dan bubur pisang. Seluruh homogenitas ragam antar perlakuan diuji dengan uji Barlett, data dikenakan analisis ragam, dan jika Vincentia Atika Septiana analisis ragam nyata maka dilakukan uji lanjut yaitu pemisahan nilai tengah dengan uji BNT pada taraf 5%. Hasil penelitian menunjukkan bahwa (1) Baik media dasar, bahan adenda organik yang ditambahkan ke dalam media maupun interaksi antar kedua faktor tersebut berpengaruh nyata terhadap pertumbuhan seedling anggrek Dendrobium in vitro, (2) media dasar Growmore menghasilkan pertumbuhan seedling Dendrobium lebih baik dibandingkan dengan media dasar ½ MS yang ditunjukkan oleh nilai rata-rata tinggi tanaman, jumlah daun, jumlah akar, panjang akar, dan bobot basah seedling jika ditambah dengan adenda ekstrak taoge. Media Growmore juga menghasilkan nilai rata-rata jumlah daun dan jumlah akar yang lebih baik daripada ½ MS jika ditambah dengan ekstrak kentang, tetapi jika ditambah dengan ekstrak tomat, maka media Growmore hanya menghasilkan nilai rata-rata jumlah akar yang lebih banyak daripada media ½ MS, (3) media ½ MS menghasilkan jumlah akar yang lebih banyak daripada media Growmore jika ditambah dengan bubur pisang ambon kuning. Abstract The objective of this research was to study effects of basal media and various organic adenda on growth of Dendrobium seedlings in vitro. The experiment was conducted in a completely randomized design with three replication. Treatments were arrange factorially with two basal media (i.e., ½ MS or Growmore) as the first factor and various organic adenda (i.e., tomato extract, mungbean sprout extract, potato extract or banana homogenate) as the second factor. Data on shoot length, number of leaves, number of roots, length of roots and shoot fresh weights were subjected to analysis of variance and if there was any significant difference among F values the analysis was followed by mean separation test using least significant difference (LSD) at P 0.05. Result of the experiment showed that (1) both basal media and various organic adenda added in each medium significantly affected growth ofDendrobium seedlings in vitro, and there was significant interaction between the two factors in affecting seedling growth. (2) Growmore basal medium resulted in better Vincentia Atika Septiana seedling growth compared to ½ MS basal medium as showed by greater values of shoot length, number of leaves, number of roots, root length, and shoot fresh weights if supplemented by mungbean sprout extract. Growmore medium also resulted in higher values of number of leaves and number of roots if supplemented by potato extract, whereas if supplemented by tomato extract, Growmore medium only resulted in higher number of roots when compared to ½ MS medium. (3) ½ MS medium resulted higher number of roots when compared to Growmore medium if supplemented by banana homogenate.
