Abstrak

Abstrak Aeromonas salmonicida merupakan bakteri pathogen penyebab penyakit carp erytrodermatitis pada ikan mas. Salah satu solusi dalam pencegahan penyakit ini adalah vaksinasi. Tujuan dari penelitian ini adalah mengetahui tingkat imunogenisitas vaksin inaktif whole cell A. salmonicida pada ikan mas serta mencari metode vaksinasi yang terbaik. Vaksin inaktif dibuat dengan cara diinaktifasi dengan formalin 1% dan diinkubasikan selama 24 jam pada suhu ruang. Vaksinasi diujikan pada 10 ekor ikan/metode vaksinasi yaitu dengan cara suntik melalui intraperitoneal (107 sel/ikan), oral (107 sel/ikan), perendaman (107 sel/ml air, lama perendaman 30 menit) dan ikan tanpa divaksinasi sebagai kontrol. Vaksinasi dilakukan sebanyak 2 kali, booster dilakukan 7 hari setelah vaksinasi I dengan dosis dan metode vaksinasi yang sama. Pengamatan titer antibodi dilakukan tiga kali yaitu, sebelum vaksinasi, 7 hari setelah vaksinasi I, dan 7 hari setelah booster. Parameter kualitas air yang diukur adalah oksigen terlarut, pH dan suhu sebagai parameter pendukung. Hasil penelitian diperoleh rata – rata titer antibodi untuk semua metode sebelum vaksinasi sebesar 1/6, setelah vaksinasi I dan booster terjadi peningkatan menjadi 1/58.67 dan 1/85.33 pada metode suntik, 1/42.67 dan 1/64 untuk metode oral, dan 1/24 dan 1/42.67 pada perendaman. Sedangkan pada ikan kontrol tanpa vaksinasi terjadi peningkatan namun tidak signifikan yaitu 1/9.33 dan 1/18.67. Metode vaksinasi terbaik adalah metode suntik secara i.p yang menunjukkan rata – rata hasil titer antibodi tertinggi. Hasil pengukuran kualitas air selama penelitian masih berada dalam kisaran normal hidup ikan uji. Kata kunci : Aeromonas salmonicida, ikan mas, titer antibodi, vaksin inaktif. Abstract Aeromonas salmonicida is bacterial pathogen that caused carp erytrodermatitis in common carp. One preventive action to protect this infection was by vaccination. The aims of this experiment were to determine the immunogenicity of inactivated vaccine A. salmonicida whole cell in common carp and the best administration method of vaccine. Vaccine was produced by inactivated A. salmonicida with 1 % formaldehyde and incubated for 24 hours in room temperature. Vaccine was tested in 10 fish/vaccination method by injection intra peritoneal (107 cell/fish), oral (107 cell/fish), immersion (107 cell/ml for 30 minute) and fish that had not vaccinated as a control. Booster was conducted in 7th days after first vaccination with same both dosage and method. Titer antibody was evaluated in three times i.e. before vaccination, 7th days after first vaccination, and 7th days after booster. Water quality such as dissolved oxygen, pH and water temperature was measured as supported parameters. The results of the experiment showed that average of titer antibody for all method before vaccination was 1/6. However, titer antibody after first vaccination and booster increased to 1/58.67 and 1/85.33 for i.p injection, 1/42.67 and 1/64 for orally, 1/24 and 1/42.67 for immerse, respectively. Beside that, there was no significantly increasing of titer antibody for fish controls that was 1/9.33 and 1/18.67. Vaccination method by i.p injection was the best method that showed the highest immunogenicity of vaccine. Water quality parameters for this experiment still in optimum range for common carp live medium. Key words: Aeromonas salmonicida, common carp, inactivated vaccine, titer antibody.
