Abstrak

Abstrak Vaksin inaktif whole cell A. salmonicida memiliki tingkat imunogenesitas yang cukup tinggi pada ikan mas (Cyprinus carpio L). Untuk menjaga stabilitas imunogenesitas vaksin tersebut, perlu dilakukan penyimpanan yang baik. Gliserol diketahui mampu menjaga keutuhan sel dalam penyimpanan dingin. Tujuan dari penelitian ini yaitu untuk mengetahui dosis gliserol terbaik dalam penyimpanan vaksin whole cell A. salmonicida.Vaksin diinaktifasi dengan formalin 1% dan diinkubasikan selama 24 jam pada suhu ruang. Kepadatan vaksin dihitung lalu ditambahkan gliserol dengan dosis 0,25%, 0,50%, 0,75% dan 2 perlakuan lain sebagai kontrol positif (vaksin tanpa gliserol) dan negatif (tanpa vaksin, berisi PBS). Vaksin diujikan pada 10 ekor ikan mas per perlakuan dengan cara penyuntikan intraperitoneal (107 sel/ikan). Seminggu berikutnya dilakukan booster dengan metode dan dosis yang sama. Vaksinasi dan booster dilakukan pada penyimpanan vaksin 0 hari dan 30 hari dengan menggunakan ikan mas yang berbeda. Titer antibodi diamati sebelum vaksinasi, seminggu setelah vaksinasi, dan seminggu setelah booster, baik pada penyimpanan 0 hari dan 30 hari. Parameter pendukung kualitas air akuarium diamati setiap hari meliputi DO, pH dan suhu. Hasil penelitian menunjukkan tingkat titer antibodi tertinggi pada vaksin dengan penambahan dosis gliserol 0,50% dengan rata-rata 256 kali pengenceran pada uji penyimpanan vaksin 30 hari. Hasil pengukuran kualitas air, secara umum masih berada dalam kisaran normal untuk kelangsungan hidup ikan mas. Kesimpulan dari penelitian ini yaitu dosis gliserol terbaik dalam mempertahankan tingkat imunogenesitas vaksin A. salmonicida adalah vaksin dengan penambahan gliserol 0,50%. To maintain the stability of the vaccine imunogenecity, the best storage was needed. Glycerol has been known to be able to maintain the integrity of the cell in cold storage. The purpose of this research was to know the best dosage of glycerol in the storage of inactive whole cell vaccine of A. salmonicida. Vaccine was inactived with 1% formaline and incubated for 24 hours at room temperature. The density of vaccine was counted and then vaccine was added with 0,25%, 0,50%, 0,75% of glycerol. Two other treatments were used as positive (vaccine without glycerol) and negative (without vaccine, contains PBS) controls. The vaccine was tested on 10 fish/treatment by intraperitoneal injection (107cell/fish). Vaccination and booster were done in 0 day and 30 days of storage using a different fish. Observation of titer antibody was done before vaccination, a week after vaccination and a week after the booster. Water quality as supporting parameter was observed every day, including DO, pH and temperature. As a results, the highest level of titer antibody was reached on vaccine with the adding dosage of 0,50% of glycerol with the average 256 times dilution on the storage of vaccine 30 days. Water quality during this research that was still in normal for common carp life. In conclusion, the best dosage to keep the level of immonugenecity of A. salmonicida vaccine was the additional 0,50% of glycerol.
