Abstrak

Abstrak Telah dilakukan isolasi dan identifikasi senyawa alkaloid dari actinomycetes ANLd-2b-3 yang hidup di lumpur hutan bakau. Senyawa isolat, A8, dimurnikan dengan metoda kromatografi kolom melalui beberapa tahapan dan diperoleh rendemen sebanyak 3.7 %. Kemurnian isolat diidentifikasi menggunakan metoda Kromatografi Cair Kinerja Tinggi (KCKT) dengan detektor Evaporating Light Scattering Detection (ELSD). Kromatogram KCKT menunjukkan satu puncak dengan nilai Retention Time (Rt) 20,6 menit. Karakterisasi dengan metoda spektroskopi Fourier Transform Infrared (FTIR) menginformasikan bahwa senyawa A8 merupakan senyawa metabolit sekunder dari golongan alkaloid. Hal ini ditunjukkan oleh adanya pita serapan C-N dari gugus amina tersier pada daerah 1109,95 cm-1 spektrum FTIR yang didukung dengan hasil uji Kromatografi Lapis Tipis (KLT) dengan pereaksi spesifik Dragendorff. Senyawa A8 juga mengandung gugus hidroksil yang ditunjukkan oleh pita serapan O-H yang melebar pada dareah 3402,33 cm-1 dan gugus karboksil pada daerah 1647,09 cm-1. Hasil uji antibakteri senyawa A8 dengan konsentrasi 5000 bpj menunjukkan bahwa senyawa A8 memiliki aktivitas antibakteri terhadap bakteri Eschericia coli dan Staphylococcus aureus. Abstract The isolation and identification of an alkaloid compound of actinomycetes ANLd-2b-3 living in mangrove mud has been carried out. The compound, A8, was purified through several stages using colum chromatography method and yielded about 3,7%. The purity of the compound was identified by High Performance Liquid Chromatography (HPLC) method using Evaporating Light Scattering Detection (ELSD) detector. The chromatogram gave a peak with Retention Time (Rt) 20,6 minute. Characterization using Fourier Transform Infra Red (FTIR) method informed that the A8 was a secondary metabolite compound of alkaloid group. It was shown by the absorption band of C-N of tertiary amine at 1109,95 cm-1 on FTIR spectrum which was also supported by Thin Layer Chromatography (TLC) test using Dragendorff reagent. The A8 compound also contained both hydroxyl group (s) shown by the absorption band at 3402,33 cm-1 and carboxyl group at1647,09 cm-1. The result of antibacterial testing with 5000 ppm concentration of A8 showed that the compound had antibacterial activities toward Eschericia coli and Staphylococcus aureus.
