Abstrak

Residu minyak pada limbah cair rumah tangga yang masuk ke perairan dapat menurunkan kualitas lingkungan. Penelitian ini ditujukkan untuk mencari mikroorganisme pendegradasi lipid limbah cair rumah tangga untuk bioremidiasi. Isolat pendegradasi lipid diisolasi dengan teknik pengenceran bertingkat. Enzim lipase dari isolat diuji dengan metode Kwon and Rhee. Dari beberapa tahapan isolasi diperoleh 1 isolat potensial, isolat bakteri tersebut mampu menghasilkan halozone disekitar koloninya. Pertumbuhan isolat yang diperoleh pada medium cair yang mengandung 2% minyak zaitun dicapai pada hari ke-3 dengan jumlah sel maksimum (OD600) sebesar 3,161. Hasil analisa 16S rDNA didapatkan bahwa isolat bakteri pendegradasi lipid adalah bakteri Pseudomonas aeruginosa sp. Hasil uji aktivitas terhadap enzim lipase isolat didapatkan nilai aktivitas unit dan spesifik enzim lipase berturut-turut sebesar 0,0841U/mL dan 0,0005U/mg. Kadar asam lemak bebas tertinggi diperoleh sebesar 0,7119 mg/mL pada waktu inkubasi hari ke-3. Kata kunci: Air limbah, bakteri pendegradasi lipid, enzim lipase ABSTRAK BAHASA INGGRIS Oil residues on household waste water that released into enviroment can decrease quality of the environment. This research was aimed to find microorganisms enable to degrade lipid in waste water. The bacteria was isolated by sequential method dilution. Lipase activity of isolates was tested using Kwon and Rhee method. From several times of isolation activities, an isolate enable generating halozone around its colony was obtained. The isolate in a liquid medium containing 2% olive oil gave the maximum number of cells (OD 600) of 3.161 for 3 days cultivation. The result of 16S rDNA analysis showed that the isolate was belong to Pseudomonas aeruginosa sp. The unit activity of the enzyme and its specific enzyme are 0.0841 U/mL and 0.0005 U/mg respectively. For 3 days incubation, 0.7119 mg/mL of free fatty acid was released to the culture. Keywords: Waste water, lipid-degrading bacteria, the enzyme lipase
