Abstrak

Anggrek tanah (Spathoglottis plicata) banyak diminati sebagai tanaman hias, ornamen taman kota, perkantoran dan kompleks perumahan di perkotaan. S. plicata dalam pertumbuhannya mengalami kendala yang cukup serius antara lain penyakit layu fusarium yang disebabkan oleh Fusarium oxysporum (Fo). Petani di Indonesia pada umumnya mengatasi penyakit layu fusarium menggunakan pestisida yang kerap menimbulkan polusi lingkungan, sedangkan tanaman anggrek selalu dekat secara fisik dengan penggemarnya. Oleh karena itu, perlu alternatif yang efektif dan ramah lingkungan. Salah satu alternatif pengendalian penyakit yang aman dan efisien terhadap lingkungan dengan menggunakan varietas tahan (resisten). Planlet anggrek tanah yang tahan terhadap Fo diseleksi secara in vitro dalam medium VW dengan penambahan asam fusarat pada berbagai konsentrasi yang berbeda. Tujuan penelitian ini untuk mengetahui: 1) Kriteria ketahanan planlet Spathoglottis plicata yang diinduksi asam fusarat terhadap infeksi F. oxysporum secara in vitro, 2) Karakter ekspresi spesifik planlet Spathoglottis plicata hasil induce resistance terhadap F. oxysporum berdasarkan aktivitas enzim peroksidase, ketebalan lignin, fenol total, kandungan klorofil total, klorofil a klorofil b, dan indeks stomata. Penelitian ini dilaksanakan di Laboratorium Botani (ruang penelitian in vitro), Jurusan Biologi, FMIPA, Universitas Lampung. Penelitian disusun menggunakan Rancangan Acak Lengkap (RAL) satu faktor yaitu konsentrasi asam fusarat terdiri atas 5 taraf : 0 ppm, 10 ppm, 20 ppm, 30 ppm, dan 40 ppm pada medium VW. Analisis data menggunakan Anova pada taraf nyata 5% dan uji lanjut dengan uji BNT (Beda Nyata Terkecil) pada taraf nyata 5%. Hasil penelitian menunjukkan bahwa: 1) Kriteria ketahanan planlet S. plicata pada hari ke-28 (0 ppm) kontrol dan 10 ppm adalah rentan. Pada 20 ppm dan 30 ppm kriteria ketahanannya yaitu moderat. Pada konsentrasi 40 ppm kriteria ketahanannya adalah tahan 2) terjadi peningkatan aktivitas enzim peroksidase, ketebalan lignin, fenol total, kandungan klorofil a, klorofil b klorofil total, dan indeks stomata planlet anggrek tanah pada berbagai konsentrasi asam fusarat Kata kunci: Spathoglottis plicata, Fusarium oxysporum, Induced Resistance, Layu Fusarium, Asam Fusarat ABSTRAK INGGRIS Ground orchid (Spathoglottis plicata) attracts many people as an ornamental plant for parks, offices and housing complexes in urban areas. In its growth, S. plicata experienced serious problems such as fusarium wilt disease caused by Fusarium oxysporum (Fo). In general, farmers in Indonesia cope with fusarium wilt disease by using pesticides that often cause environmental pollution, while orchids are always physically close to the fans. Therefore, it should be an effective and environmentally friendly alternative. One of the most secure and efficient alternative diseases control for environment is by using resistant varieties. Ground orchid plantlets that are resistant to Fo have been selected by in vitro selection on VW medium containing fusaric acid at different concentrations. The objectives of this research were to determine: 1) Resistance criteria of Spathoglottis plicata plantlet that was induced by fusaric acid on F. oxysporum infection through in vitro selection, 2) Specific expression character of Spathoglottis plicata plantlet from induced resistant to fusaric acid on F. oxysporum based on the activities of peroxidase enzyme, lignin thickness, total phenols, total chlorophyll content, chlorophyll a and b, and stomatal index. This research was conducted at Botany Laboratory (in vitro study room) of Biology Department of Mathematics and Science Faculty of Lampung University. The research was compiled by using Completely Randomized Design (CRD) with one factor that is fusaric acid consisting of 5 levels: 0 ppm, 10 ppm, 20 ppm, 30 ppm, and 40 ppm on WV medium. Data analysis used ANOVA (analysis of varian) at significance level 5% and a further test with LSD (Least Significant Difference) at the significance level 5%. The result showed that: 1) The resistance criteria of S. plicata plantlet on day 28 (0 ppm) was control and 10 ppm was susceptible. At 20 ppm and 30 ppm, its resistance criteria were moderate. At concentration of 40 ppm, its resistance criteria was resistant. 2) There was an increase in activities of peroxidase enzyme, lignin thickness, total phenols, total chlorophyll content, chlorophyll a and b, and stomatal index of ground orchid at different concentration of fusaric acid.
