Abstrak

ABSTRAK Protease banyak dimanfaatkan diberbagai bidang diantaranya industri, kesehatan maupun peternakan. Bakteri dari genus Bacillus telah banyak dikembangkan untuk produksi enzim protease dan memiliki keuntungan tersendiri. Faktor lingkungan seperti medan magnet dan ion logam dapat mempengaruhi aktivitas Bacillus sp. dalam menghasilkan enzim. Fe dan Zn adalah salah satu ion logam yang dapat berperan sebagai induktor maupun inhibitor pada pertumbuhan dan aktivitas Bacillus sp. dalam menghasilkan enzim protease. Tujuan penelitian ini adalah: 1) Mengetahui pengaruh paparan medan magnet 0.2 mT pada ion logam Fe dan Zn terhadap aktivitas Bacillus sp. dalam menghasilkan enzim protease. 2) Membedakan pengaruh paparan medan magnet 0.2 mT antara ion logam Fe dan Zn terhadap aktivitas enzim protease Bacillus sp. Penelitian ini dilakukan menggunakan secara bertahap. Tahap pertama meliputi isolasi dan seleksi bakteri. Tahap kedua yaitu uji proteolitik pada media padat dengan 6 perlakuan yaitu M0L0, M1L0, M0L1, M1L1, M0L2 dan M1L2. Tahap ketiga meliputi produksi enzim potease pada media cair dengan 4 perlakuan yaitu M0L0, M1L0, M0L1, M1L1. Hasil penelitian menunjukkan bahwa: 1) Paparan medan magnet 0.2 mT selama 10 menit pada ion logam Fe dan Zn berpengaruh terhadap aktivitas Bacillus sp. dalam menghasilkan enzim protease. 2) Paparan medan magnet 0.2 mT selama 10 menit pada ion logam Fe meningkatkan aktivitas enzim dan jumlah sel hidup Bacillus sp. sedangkan paparan medan magnet 0.2 mT selama 10 menit pada ion logam Zn menghambat aktivitas enzim dan pertumbuhan Bacillus sp. Kata kunci : Protease, Bacillus sp., Medan Magnet, Ion Logam Fe dan Zn. ABSTRACT Protease has been widely used in various fields including industry field, health and animal husbandry. Bacteria of the genus Bacillus have been developed to produce protease and have many advantages. The environmental factors such as magnetic field can also affect the activity of Bacillus sp. to produce enzymes. Fe atoms are one of the ferromagnetic compounds that are used as an inductor on Bacillus sp growth and activity in producing protease while Zn atoms, that are one of diamagnetic group, can be a protease inhibitors. The objectives of this research were: 1) Determine the effect of 0.2 mT magnetic field exposure of Fe and Zn metal ions on its induction power in the activity of Bacillus sp. to produce protease. 2) Distinguish the effect of 0.2 mT magnetic field exposure of Fe and Zn metal ions on the activity of the protease by using Bacillus sp. This research was carried out in stages. The first phase included the isolation and selection of bacteria. The second stage was the proteolysis test on solid media with 6 treatments, M0L0, M1L0, M0L1, M1L1, M0L2 and M1L2. The third phase included the production of protease on liquid media with 4 treatments, M0L0, M1L0, M0L1, and M1L1. The results showed that: 1) The exposure of 0.2 mT magnetic field 0.2 for 10 minutes at Fe and Zn metal ions affected the activity of Bacillus sp. to produce protease. 2) The exposure of 0.2 mT magnetic field for 10 minutes at Fe metal ions increased the enzyme activities and living cells of Bacillus sp. whereas the exposure of 0.2 mT magnetic field for 10 minutes at Zn metal ions inhibited the enzyme activity and the growth of Bacillus sp.
