Abstrak

ABSTRAK Produksi buah sirsak di Indonesia terus mengalami penurunan. Salah satu hama penyebabnya adalah kutu putih (P. cryptus). Penggunaan insektisida sintetik berdampak negatif, maka dibutuhkan insektisida nabati yang berasal dari tanaman yaitu gamal (G. maculata) yang mengandung bahan aktif senyawa flavonoid. Penelitian dilakukan pada bulan April – Juni 2016 di Laboratorium Zoologi dan Laboratorium Sentra Terpadu Inovasi Teknologi Universitas Lampung. Penelitian bertujuan untuk mengetahui perbandingan daya racun isolat murni ekstrak metanol dan ekstrak air daun gamal terhadap mortalitas kutu putih. Ekstraksi dilakukan dengan cara maserasi bertingkat. Fraksinasi, penapisan dan pemurnian senyawa flavonoid ekstrak polar (metanol dan air) serbuk daun gamal dan uji daya insektisidanya pada skala Laboratorium terhadap P. cryptus. Bioassay dilakukan terhadap kutu putih pada buah sirsak yang sudah direndam dalam ekstrak polar serbuk daun gamal selama 10 menit pada tingkatan konsentrasi 0%, 0,02%, 0,04%, 0,06% dan 0,08%, percobaan dilakukan 3 kali ulangan, dan diamati pada 12, 24, 48 dan 72 jam setelah perlakuan. Analisis data menggunakan probit untuk menentukan nilai LC50, uji anara dan uji lanjut Tukey’s. Hasil penelitian menunjukkan bahwa ekstrak metanol dan ekstrak air daun gamal efektif terhadap mortalitas hama kutu putih. LC50 pada 72 jam ekstrak murni metanol sebesar 0,096% dan LT50 sebesar 95,876 jam. dan LC50 Ekstrak murni air adalah 0,061% dan LT50 sebesar 69,296 jam. Dengan demikian ekstrak murni air lebih cepat dan lebih efektif daripada ekstrak metanol. Kata kunci: ekstrak metanol, ekstrak air, P. cryptus, A. muricata, G. maculata ABSTACT Soursop production in Indonesia continues to decline from year to year. One cause is the mealybug pest (Pseudococcus cryptus). the mealybug suck the young fruits soursop to dry and stunted. To control the pest, using botanical insecticides more safety than synthetic insecticides. One of the plants that can be used is Gliricidia maculata. The leaves of G. maculata contain planty of an active compound flavonoid. The study conducted in April – June 2016 in the Laboratory of Zoologi and an Integrated Center For Technological Innovation Laboratory of Lampung University. The purpose of the study to compare the toxicity of the purified isolates of water and methanol extracts of G. maculata leaves on mortality soursop mealybugs (P. cryptus). Extraction was done by maceration series using various organic solvents (n-hexane, dichloromethane, methanol and water). Fractionation and purification of flavonoids from polar extracts were done by Chromatography Coloum. A set of laboratory expriment was conducted by using block design. Methanol and Water extracs (ME and WE) with 5 levels concentration i.e. 0%, 0.02%, 0.04%, 0.06% and 0.08%, and 3 replications. ANOVA was conducted to obtain the means and standard deviations of the exprimental study, and Tukey’s test at α = 5% was peformed in order to obtain the different among the exprimental groups. Analisys Probit were used for compare the efectivities the exstracts. The result indicated the was toxic to mealybug pest (P. cryptus) with LC50 72 hours metanol extract 0,096% and LT50 metanol extract 95, 876 hours and water extract 0,061% and LT50 69,296 hours.
