Abstrak

Indonesia merupakan produsen rumput laut nomor 1 dunia. Rumput laut Eucheuma cottonii merupakan salah satu jenis rumput laut penghasil kappa karaginan yang mengandung lebih dari 34% 3,6 anhidro-D-galaktosa dan 25% ester sulfat. Rumput laut Eucheuma cottonii dapat menghasilkan karaginan dengan cara ekstraksi. Ekstraksi rumput laut dipengaruhi oleh pelarut alkali, suhu ekstraksi, dan pengendap. Tujuan penelitian ini yaitu untuk mengetahui pengaruh tiga variabel bebas (konsentrasi NaOH, suhu ekstraksi dan konsentrasi KCl) terhadap rendemen, sifat fisik, dan kimia karaginan murni, serta mendapatkan kondisi optimum proses ekstraksi rumput laut Eucheuma cottonii menjadi karaginan murni dan rendemen bermutu tinggi. Rancangan percobaan yang digunakan untuk menentukan kondisi optimum masing-masing variabel adalah metode respon permukaan (Response Surface Methodology atau RSM). Penelitian ini menggunakan tiga faktor yaitu konsentrasi NaOH (0,6N, 0,9N, dan iv 1,2N), suhu ekstraksi (80OC, 85OC, dan 90OC) dan konsentrasi KCl (0,4%, 0,6%, dan 0,8%). Setelah ekstraksi, karaginan murni dianalisis kekuatan gel, viskositas, kadar abu, kadar abu tidak larut asam, dan kadar sulfat. Hasil penelitian menunjukkan bahwa konsentrasi NaOH berpengaruh pada semua parameter selain kadar abu tidak larut asam; konsentrasi pengendap KCl berpengaruh nyata pada semua parameter selain kekuatan gel; dan suhu ekstraksi hanya berpengaruh pada kekuatan gel, viskositas, dan kadar abu tidak larut asam. Kondisi optimum terjadi apabila ekstraksi rumput laut menggunakan konsentrasi pelarut NaOH 1,5N pada suhu 93OC dengan konsentrasi pengendap KCl 0,7% dapat menghasilkkan rendemen karaginan murni maksimal yaitu sebesar 68,82%. Karakteristik mutu karaginan murni yang dihasilkan yaitu kekuatan gel 613,87 gram/cm2, viskositas 5,08 cps, kadar abu 15,38%, kadar abu tidak larut asam 2,15%, dan kadar sulfat 35,16%. Karaginan murni ini memenuhi standar mutu FAO. Eucheuma cottonii is one type of seaweed containing kappa carrageenan which contains more than 34% anhidro 3,6-D-galactose and 25% ester sulfate. Extraction of refined carrageenan from Eucheuma cottonii is affected by several variables. The objectives of this research were (1) to determine the effects of three independent variables (NaOH concentration, temperature extraction and KCl concentration) on the yield and properties of refined carrageenan, as well as (2) to find out the optimum conditions of refined carrageenan extraction which was able to result in the highest yield. To achieve the objectives, Response Surface Methodology (RSM) experimental design was implement in this research with three free variable, namely concentrations of NaOH (0,6N, 0,9N, and 1,2N), the temperatures of extraction (80oC, 85oC, and 90oC) and concentration of KCl (0,4%, 0,6%, and 0,8%). After extraction, the refined carrageenan was analyzed its gel strength, ii viscosity, ash content, acid insoluble ash content, and sulfate content. The research results showed that (1) the NaOH concentration affected significantly on all parameters except on its acid in soluble ash content; KCl concentration also affected significantly on all parameters except on its gel strength; extraction temperatures only affected significantly on gel strength, viscosity, and acid insoluble ash content; and (2) the optimum condition of refined carrageenan extraction occurred at the NaOH concentration of 1.5 N, the extraction temperature of 93oC, and the KCl concentration of 0.7%. The refined carrageenan yield at the optimum extraction condition was 68.82% and it properties as follow: 613.87 g/cm2 gel strength, 5.08 cPs viscosity, 15.38% ash content, 2.15% acid insoluble ash content, and 35.16% sulfate content. These properties fulfilled the refined carrageenan quality standard of FAO. Keywords: Eucheuma cottonii seaweed, extraction, KCl, refined carrageenan, response surface methodology (RSM).
