Abstrak

ABSTRAK ( Bahasa Indonesia) Limbah cair nanas mengandung bahan organik diantaranya amilum. Amilum dapat dihidrolisis oleh mikroorganisme amilolitik. Berdasarkan hasil penelitian sebelumnya,diperoleh bakteri indigen dari limbah cair nanas yang memiliki kemampuan untuk mengdirolisis amilum diantaranya Bacillus cereus, dan Bacillus substilis. Tujuan dari pelitian ini adalah untuk mengetahui karakter fisiologis dan indeks amilolitik bakteri Bacillus dari limbah cair nanas. Rancangan percobaan yang digunakan dalam penelitian ini yaitu Rancangan Acak kelompok (RAK) dengan tiga kali pengulangan. Data yang diperoleh dianalisis ragam, kemudian diuji lanjut dengan Uji Beda Nyata Terkecil (BNT) pada taraf α 5 %. Hasil penelitian diperoleh 5 isolat bakteri Bacillus amilolitik yang memiliki kesamaan sifat yaitu bersifat Gram positif, katalase positif, motil, tidak dapat mencairkan gelatin namun dalam berbeda kemampuannya dalam memfermentasikan gula-gula. Indeks amilolitik yang terbaik adalah isolat Bacillus sp.5 dengan nilai indeks 5,146, tergolong bakteri amilolitik dengan aktivitas tinggi, diikuti isolat Bacillus sp.4 sebesar 4,416 dan Bacillus sp.3 sebesar 4,236, sedangkan indeks amilolitik yang sedang adalah isolat Bacillus sp.2 nilai indeks sebesar 1,216, isolat Bacillus sp.1 sebesar 2,609. Penentuan indeks amilolitik yang tertinggi di dapat pada hari ke-3 dengan nilai indeks 5,146. Starch can be hydrolyzed by amylolytic microorganisms. Based on the research results previous derived from the bacterium indigen pineapple liquid waste that has the ability to hydrolysis starch include Bacillus cereus, and Bacillus substilis. The purpose of this research is to know the characters and physiological indexes of amylolytic bacteria Bacillus from pineapple liquid waste. The experimental design used in this study is a group Randomized Block Design (RBD) with three replications. Data were analyzed for variance, then tested further by Least Significant Difference Test (LSD) at α level of 5%. The results were obtained 5 amylolytic Bacillus isolates that have similar properties which are Gram-positive, catalase positive, motile, can not liquefy gelatin, but different in its ability to ferment sugars. Index is best amylolytic Bacillus isolates sp.5 with an index value of 5.146, amylolytic bacteria categorized with high activity, followed by 4.416 sp.4 Bacillus isolates and Bacillus sp.3 amounted to 4,236, while the index is being amylolytic Bacillus isolates sp.2 value index of 1.216, 2.609 isolates of Bacillus sp.1. Determination of the highest amylolytic index can be on day 3 with an index value of 5.146. Keywords: pineapple liquid waste, exploration, amylolytic, Bacillus, and starch
