Abstrak

Penelitian ini bertujuan untuk mempelajari pengaruh penambahan sorbitol terhadap kestabilan enzim selulase dari Rhizopus oligosporus menggunakan senyawa poliol yaitu sorbitol. Adapun tahapan yang dilakukan dalam penelitian ini meliputi : produksi, isolasi, pemurnian, penambahan zat aditif menggunakan sorbitol dengan konsentrasi 0,5; 1; 1,5 M dan karakterisasi enzim selulase sebelum dan setelah penambahan sorbitol. Hasil penelitian menunjukkan bahwa aktivitas spesifik enzim selulase tanpa sorbitol 35,2038 U/mg, meningkat 10 kali dibandingkan dengan ekstrak kasar enzim yang mempunyai aktivitas spesifik 3,48078 U/mg. Enzim selulase tanpa sorbitol mempunyai pH optimum 5,5; suhu optimum 65oC; KM = 18,69 mg/mL substrat; Vmaks = 0,856 μmol/mL.menit. Uji stabilitas termal selama 60 menit pada suhu 65oC enzim selulase mempunyai nilai ki = 0,045 menit-1; waktu paruh (t1/2) = 15,40 menit dan ΔGi = 98,65 kJ/mol. Enzim setelah penambahan sorbitol mempunyai pH dan suhu optimum yang sama dengan enzim tanpa sorbitol. Data kinetika enzim setelah penambahan sorbitol 0,5 M diperoleh KM = 17,37 mg/mL, substrat dan Vmaks = 0,596 μmol/mL.menit, t1/2 = 19,80 menit, ki = 0,035 menit dan ΔGi = 99,32 kJ/mol. Data kinetika enzim setelah penambahan sorbitol 1 M diperoleh KM = 14,72 mg/mL, substrat dan Vmaks = 0,342 μmol/mL.menit, t1/2 = 22,35 menit, ki = 0,031 menit dan ΔGi = 99,65 kJ/mol. Data kinetika enzim setelah penambahan sorbitol 1,5 M diperoleh KM = 11,33 mg/mL, substrat dan Vmaks = 0,223 μmol/mL.menit, t1/2 = 27,72 menit, ki = 0,025 menit dan ΔGi = 100,23 kJ/mol. Berdasarkan penurunan nilai ki, peningkatan waktu paruh (t1/2) dan nilai ΔGi, diketahui bahwa enzim setelah ditambahkan senyawa aditif menggunakan sorbitol dapat meningkatkan stabilitas enzim selulase dari Rhizopus oligosporus. Kata kunci : Rhizopus oligosporus, selulase, zat aditif, sorbitol abstract This research was conducted to determine the stability of the enzyme before and after the addition of sorbitol compound cellulase enzyme from Rhizopus oligosporus using the polyol compound sorbitol. Scope of this reseach were including production, isolation, purification, addition of additives using sorbitol with a concentration of 0.5; 1.0; 1.5 M and characterization of purified cellulase enzymes before and after sorbitol addition. The result showed that the purified enzyme has a specific activity 35,2038 U/mg or increase for 10 times than crude extract enzyme which has specific activity 3,48078 U/mg respectively. The purified enzyme have some characteristics are optimum reaction reach at pH 5,5; temperature 65oC; KM = 18,69 mg/mL; Vmax = 0,856 μmol/mL.min. A thermal stability test for 60 min at a temperature of 65oC of cellulase enzyme has ki = 0,043 min-1; t1/2 = 16,11 min and ΔGi = 98,77 kJ/mol. The enzyme after addition of sorbitol has the same pH and temperature as the purified enzyme. Enzyme kinetics data after addition of sorbitol 0.5 M were obtained KM = 17,37 mg/mL; Vmax = 0,596 μmol/mL.min; ki = 0,039 min-1; t1/2 = 17,76 min and ΔGi = 99,32 kJ.mol-1. Enzyme kinetics data after addition of sorbitol 1 M were obtained KM = 14,72 mg/mL; Vmax = 0,342 μmol/mL.min; ki = 0,031 min-1; t1/2 = 22,35 min and ΔGi = 99,65 kJ/mol. Enzyme kinetics data after addition of sorbitol 1,5 M were obtained KM = 11,32 mg/mL; Vmax = 0,223 μmol/mL.min; ki = 0,029 min-1; t1/2 = 25,66 min and ΔGi = 100,23 kJ/mol. Based on the decrease of ki values, increase of t1/2 and ΔGi, are known that enzyme after addition sorbitol can increase the stability of cellulase from Rhizopus oligosporus.
