Abstrak

Selulase banyak digunakan dalam industri untuk mengkonversi selulosa menjadi glukosa. Agar dapat digunakan dalam proses industri, diperlukan enzim yang stabil pada pH dan suhu ekstrim. Penelitian ini bertujuan untuk meningkatkan stabilitas enzim selulase dari isolat bakteri Bacillus subtilis ITBCCB148 melalui proses amobilisasi enzim menggunakan bentonit. Tahapan yang dilakukan dalam penelitian ini adalah produksi, isolasi, pemurnian, amobilisasi dan karakterisasi enzim selulase hasil pemurnian dan hasil amobilisasi. Hasil penelitian menunjukkan aktivitas spesifik enzim hasil pemurnian sebesar 14,075 U/mg, meningkat kemurniannya 5,7 kali dibandingkan ekstrak kasar enzim. Enzim selulase hasil pemurnian memilik i suhu optimum 50ºC, sedangkan enzim amobil pada suhu 60ºC. Uji stabilitas termal pada suhu 60ºC selama 100 menit untuk enzim hasil pemurnian masih memiliki aktivitas sisa 7,330 %, sedangkan enzim amobil sebesar 14,681%. Data kinetika enzim hasil pemurnian diperoleh KM = 1,278 mg mL-1 substrat dan Vmaks = 1,271 μmol mL-1 menit-1, t1/2 = 31,111 menit, ki = 0,022 menit-1 dan ΔGi =100,540 KJ mol-1. Pada data kinetika enzim hasil amobilisasi diperoleh KM = 0,445 mg mL-1 substrat dan Vmaks = 0,422 μmol Ml menit-1, t1/2 = 39,833 menit, ki = 0,017 menit-1 dan ΔGi = 104,424 KJ mol-1. Berdasarkan penurunan nilai ki , peningkatan waktu paruh (t1/2) dan nilai ΔGi, diketahui bahwa amobilisasi menggunakan bentonit dapat meningkatkan stabilitas enzim selulase dari Bacillus subtilis ITBCCB148. For a certain industrial processes, the enzyme must be set up at extreme level of both pH and temperature. This research to increase the stability of cellulose enzyme from Bacillus Subtilis ITBCCB148 by the enzyme immobilization process using bentonite. The steps in this research are: production, isolation, purification, immobilization and characterization of purified and immobilized enzymes. The research show the specific activity of the puried enzyme of 14,075 U/mg, increased of 5,7 times than the crude extract. The purified cellulase enzyme has an optimum temperature at 500C. Whereas the immobilized enzyme at 600C. The residual activity on 600C for 100 minutes for purified enzyme was 7.330%, while the immobilized enzyme was 14.681%. Kinetic datas of purified enzyme results were KM = 1.278 mg mL-1 substrate, Vmax = 1.271 µmol mL-1 minute-1, t1/2 = 31.111 minutes, Ki = 0.022 min-1 and ΔGi = 100.540 KJ mol-1. The datas of the immobilized enzyme were KM = 0.445 mg mL-1 substrate, Vmax = 0.442 µmol mL-1 minute-1, t1/2 = 39.833 minutes, Ki = 0.017 min-1 and ΔGi = 104.424 KJ mol -1. Based on the decrease of ki, increase of half-time (t½) and the value of ΔGi the immobilization by using bentonite can improve the stability of cellulose enzyme from Bacillus Subtilis ITBCCB148.
