Abstrak

PENAPISAN BAKTERI PENGHASIL ENZIM AMILASE DARI USUS IKAN GURAME (Osprhonemus gouramy) Oleh GUSNITA ALFIN Ikan gurame (Osprhonemus gouramy) merupakan ikan herbivora yang pertumbuhannya lambat. Upaya untuk meningkatkan pertumbuhan yaitu dengan meningkatkan kinerja pencernaannya. Enzim yang berperan penting dalam pencernaan ikan herbivora adalah enzim amilase. Enzim amilase dapat memecah beberapa sumber karbohidrat sehingga mudah diserap oleh tubuh. Peningkatkan aktivitas enzim amilase dapat dilakukan dengan memanfaatkan bakteri indigeneous dari usus ikan gurame yang dapat menghasilkan enzim amilase. Tujuan dari penelitian ini adalah untuk mendapatkan isolat bakteri penghasil enzim amilase yang mampu memecah karbohidrat. Terdapat 102 isolat dari 15 sampel ikan dari 5 lokasi yang berbeda di Lampung. Satu isolat (D.2) potensial memecah tepung terigu sebagai sumber karbohidrat. Hal ini ditunjukkan dengan adanya zona bening terhadap media TSA yang telah ditambahkan tepung terigu. Hasil identifikasi dengan pengamatan morfologi dan uji biokimiawi menunjukkan isolat D.2 sebagai bakteri Pseudomonas cepacia. Kata Kunci : Indigeneous, amilase, usus gurame ABSTRACT SCREENING OF BACTERIA PRODUCING AMYLASE ENZYME FROM THE INTESTINES OF GURAME (Osprhonemus gouramy) By GUSNITA ALFIN Gurame (Osphronemus gouramy) is a slow growing herbivorous fish. The effort to improve growth performance is by fostering digestion. Amylase is an enzyme that played an important role in the digestion of herbivorous fish. Amylase can hydrolized carbohydrate sources in gurame digestion to be easily absorbed. Amylase activity boosting can be done by utilizing the intestinal bacteria of indigeneous gurame that can produce the amylase. The purpose of this research was isolat bacteria producing amylase capable of hydrolized carbohydrates. The result of the research we isolated 102 bacteria from 15 gouramy in 5 different locations in Lampung. One isolate (D.2) capable to hydrolized wheat flour. It was indicated by the presence of a clear zone on the TSA that had been added to wheat flour to test the potential of amylase. The identification by observation morphology and biochemical tests show isolates (D.2) as bacterium Pseudomonas cepacia. Keywords: Screening, gurame, indigenous, amylase, digestion
