Penelitian ini bertujuan untuk mempelajari pengaruh infestasi trematoda terhadap leukosit dan diferensial leukosit darah sapi Simpo. Penelitian dilaksanakan pada Desember 2018 sampai Januari 2019, bertempat di Desa Labuhan Ratu, Kecamatan Labuhan Ratu, Kabupaten Lampung Timur. Pemeriksaan leukosit dan diferensialnya dilaksanakan di Balai Veteriner Lampung. Rancangan percobaan yang digunakan adalah Rancangan Acak Lengkap dengan 3 perlakuan dan 4 kali ulangan. Perlakuan yang digunakan adalah P0: sapi Simpo yang tidak terinfestasi trematoda; P1: sapi Simpo yang terinfestasi satu jenis trematoda; P2: sapi Simpo yang terinfestasi dua jenis trematoda. Data hasil pengamatan dianalisis dengan sidik ragam pada taraf nyata 5%. Hasil penelitian ini menunjukkan bahwa infestasi trematoda tidak berpengaruh nyata (P>0,05) terhadap leukosit dan diferensialnya. Rata-rata nilai leukosit darah sapi Simpo masih berada dalam kisaran normal dengan nilai tertinggi pada P2 (11,43 × 103 /μL) dan terendah pada P1 (10,80 × 103 /μL), sedangkan diferensial leukosit yang melebihi batas normal yaitu monosit pada P0 (1,99 × 103 /μL) dan eosinofil pada P1 (1,22 × 103 /μL). Diferensial leukosit yang berada dibawah kisaran normal yaitu netrofil pada P0 (1,39 × 103 /μL). Kata kunci: Leukosit, Diferensial leukosit, Trematoda, Sapi simpo. This research aims to study the effect of trematode infestation on leukocyte and its differential in Simpo cattle blood. The research was conducted on December 2018 until January 2019, located in Labuhan Ratu Village, Labuhan Ratu District, East Lampung Regency. Leukocyte and its differential examination are carried out at Lampung Veterinary Center. The experimental design was used Completely Randomized Design with 3 treatments and 4 replications. The treatments were used P0: non infested Simpo cattle; P1: Simpo cattle infested with one type of trematode; P2: Simpo cattle infested with two types of trematode. The observed data were analyzed by analysis of variant with the assumptions 5%. The results showed that trematode infestation had no effect (P> 0.05) on leukocyte and its differential. The average value of leukocyte were still within the normal range with the highest value on P2 (11.43 × 103 / μL) and the lowest value on P1 (10.80 × 103 / μL), while the leukocyte differential that exceed the normal range were found in monocyte on P0 (1.99 × 103 / μL) and eosinophil on P1 (1.22 × 103 / μL). The leukocyte differential that below the normal range was found in neutrophil on P0 (1,39 × 103/μL). Keywords: Leukocyte, Differential leukocyte, Trematode, Simpo cattle.
