Abstrak

Resistensi bakteri terhadap antibiotik menjadi permasalahan global dan menimbulkan kebutuhan mendesak untuk mencari senyawa metabolit baru dari bahan alam. Ekosistem mangrove memiliki karakteristik lingkungan yang unik sehingga mikroba yang mampu bertahan dalam kondisi tersebut memiliki bioaktivitas salah satunya sebagai antibakteri. Pada penelitian ini sedimen mangrove diambil secara acak di kawasan ekowisata hutan mangrove Gebang Petengoran Kabupaten Pesawaran dengan titik koordinat -5.570759°LS 105.240765°BT, dan kawasan pantai Dewi Mandapa dengan titik koordinat -5.571883°LS 105.243494°BT. Hasil isolasi diperoleh empat isolat fungi yang difermentasi menggunakan media beras padat. Isolat terpilih RSM1 dengan indikasi genus Rhizidium, ekstrak hasil ko-kultivasi menunjukkan nilai % inhibisi sebesar 21.2% terhadap Staphylococcus aureus dan 26.8% terhadap Pseudomonas aeruginosa. Analisis FTIR menunjukkan adanya serapan pada bilangan gelombang 1371.7 cm-1 menunjukkan serapan gugus C-N yang merupakan gugus fungsi senyawa alkaloid. Serapan pada bilangan gelombang 1744,4 cm-1 dan 2922.2 cm-1 menunjukkan adanya serapan C=O dan C-C alifatik. Analisis LC-MS senyawa fraksi RSM1 diperoleh komponen alkaloid berupa best peak dengan m/z 365 dengan formula molekul C17H33N8O hasil analisis mengindikasi struktur alkaloid dengan kerangka dasar triazine. Kata kunci : Sedimen mangrove, fungi, antibakteri, alkaloid. ABSTRACT Bacterial resistance to antibiotics is a global problem and creates an urgent need to look for new metabolites from natural ingredients. Mangrove ecosystems have unique environmental characteristics so that microbes that are able to survive in these conditions have bioactivity, one of which is antibacterial. In this study, mangrove sediments were taken randomly in the ecotourism area of the Gebang Petengoran mangrove forest, Pesawaran Regency with the coordinates of -5.570759°S 105.240765°E, and the Dewi Mandapa beach area with the coordinates of -5.571883°S 105.243494°E. The results of the isolation obtained four isolates of fungi that were fermented using solid rice media. The selected isolate RSM1 with an indication of the genus Rhizidium, the extract from the co-cultivation method an inhibition value of 21.2% against Staphylococcus aureus and 26.8% against Pseudomonas aeruginosa. FT-IR analysis showed an absorption at wave number of 1371.7 cm-1 indicating the absorption of the C-N group which is a functional group of alkaloid compounds. Absorption at wave numbers 1744.4 cm-1 and 2922.2 cm-1 indicates aliphatic C=O and C-C absorption. LC-MS analysis of RSM1 fraction compounds obtained alkaloid components in the form of the best peak with m/z 365 with the molecular formula C17H33N8O. The results of the analysis indicated the structure of the alkaloids with base structure of triazine. Key words : Mangrove sediment, fungi, antibacterial, alkaloid.
