Abstrak

Produksi bacterial nanocellulose (BNC) umumnya menggunakan glukosa sebagai sumber karbon pada medium Hestrin-Schramm (HS) cair. Limbah nanas berpotensi sebagai sumber karbon alami dalam media fermentasi untuk produksi BNC karena mengadung karbohidrat sebesar 17,53%. Penggunaan limbah nanas sebagai sumber karbon alternatif pengganti glukosa diharapkan meminimalisir biaya produksi BNC. Penelitian ini bertujuan untuk memperoleh isolat bakteri penghasil BNC dari kombucha dan memperoleh kondisi optimum produksi BNC menggunakan sumber karbon dari limbah nanas. Produksi BNC dilakukan dengan menggunakan medium HS cair yang dimodifikasi glukosanya menggunakan limbah nanas dengan variasi konsentrasi 2%, 4%, 6%, dan 8 %. Produksi BNC dilakukan dengan variasi keadaan fermentasi (statis dan agitasi dengan kecepatan 150 rpm) dan variasi waktu inkubasi (7, 14, dan 21 hari). Parameter yang diukur meliputi berat pelikel BNC dan Water Hold Capacity (WHC). Pelikel BNC yang diperoleh pada kondisi optimum dikarakterisasi menggunakan SEM (Scanning Electron Microscopy). Hasil penelitian diperoleh isolat Kc-D-4 yang mampu menghasilkan BNC. Produksi BNC oleh isolat Kc-D-4 pada sumber karbon limbah nanas optimum pada konsentrasi 8% dan waktu inkubasi 14 hari dalam keadaan statis. Pada kondisi tersebut dihasilkan berat basah pelikel BNC sebesar 10,2 g dengan WHC sebesar 97,30%. Hasil karakterisasi BNC menggunakan SEM menunjukkan morfologi struktur jaringan serat saling berhubungan dan rapat serta memiliki serat berukuran nano dengan diameter 39-65 nm. However, the pineapple waste can be a natural carbon source in the fermentation medium for BNC production because it contains 17.53% carbohydrates. The use of pineapple waste as an alternative source of carbon in place of glucose is expected to minimize the cost of BNC production. This study is aimed to obtain isolates of BNC-producing bacteria from kombucha and obtain optimum conditions for BNC production using carbon sources from pineapple waste. BNC production was carried out using a modified liquid HS medium using pineapple waste with variations in concentrations of 2%, 4%, 6%, and 8%. In addition, BNC production was carried out with variations in fermentation conditions (static and agitate at a speed of 150 rpm) and variations in incubation time (7, 14, and 21 days). The parameters measured include the BNC's pellicle weight and Water Hold Capacity (WHC). BNC pellicle obtained under optimum conditions was characterized using SEM (Scanning Electron Microscopy). The results of the study obtained Kc-D-4 isolates capable of producing BNC. Production of BNC by Kc-D-4 isolates at the optimum pineapple waste carbon source at a concentration of 8% and an incubation time of 14 days in a static state. In these conditions, the wet weight of BNC is produced at 10.2 g with a WHC of 97.30%. The results of BNC characterization using SEM showed the morphology of the fiber network structure is interconnected and tight and has nano-sized fibers with a diameter of 39-65 nm.
