The use of antibiotics in fish farming activities is carried out to prevent and treat fish diseases caused by bacteria. Continuous excessive and inappropriate use of antibiotics in fish farming activities causes bacteria to become resistant. One of the bacteria reported to be resistant to antibiotics due to excessive and inappropri- ate use in fish farming activities such a Escherichia coli. The purpose of this study was to identify E. coli in freshwater fish cultivation water in Banten Province and evaluate the sensitivity of E. coli to three types of antibiotics, namely tetracycline, oxytetracycline, and enrofloxacin. E. coli isolates were identified using biochemi- cal tests using a vitek 2 compact, then the sensitivity test of E. coli to three types of antibiotics was carried out in vitro using the diffusion method (disk test). E. co- li isolates that were resistant to certain antibiotics were continued with the mini- mum inhibitory concentration (MIC) test. Test results were interpreted according to the Clinical Laboratory Standards Institute (CLSI). Based on the research re- sults, 45 E. coli isolates were obtained with a diffusion test resistance rate of 33.33% for tetracycline, 35.56% for oxytetracycline, and 15.55% for enrofloxacin. It was known that 16 E. coli isolates were resistant. Fifteen isolates resistant to te- tracycline had different MIC values, including 2 isolates had MIC values >256 μg/mL, 11 isolates had MIC values of 256 μg/mL and 2 isolates had MIC values of 128 μg/mL. Sixteen isolates experienced resistance to oxytetracycline had di- fferent MIC values, including 10 isolates having an MIC value >256 μg/mL, 5 isolates having an MIC value of 256 μg/mL and 1 isolate having an MIC value of 128 μg/mL. Seven isolates experienced resistance to enrofloxacin had different MIC values, including 3 isolates had an MIC value of 128 μg/mL, 3 isolates had an MIC value of 64 μg/mL and 1 isolate had an MIC value of 16 μg/mL. Key words: enrofloxacine, Escherichia coli, oxytetracycline, resistance, tetracycline ABSTRAK KONTAMINASI Escherichia coli (ESCHERICH, 1885) PADA AIR BUDI DAYA IKAN AIR TAWAR DI PROVINSI BANTEN : IDENTIFIKASI DAN UJI KEPEKAAN TERHADAP TIGA JENIS ANTIBIOTIK Oleh Muthia Angelia Penggunaan antibiotik pada kegiatan budi daya ikan dilakukan untuk pencegahan serta pengobatan penyakit ikan yang disebabkan oleh bakteri. Penggunaan antibi- otik secara berlebihan dan tidak tepat secara terus - menerus pada kegiatan budi daya ikan menyebabkan bakteri menjadi resisten. Salah satu bakteri yang dilapor- kan mengalami resisten terhadap antibiotik akibat penggunaan secara berlebihan dan tidak tepat pada kegiatan budi daya ikan yaitu Escherichia coli. Tujuan dari penelitian ini untuk mengidentifikasi E. coli pada air budi daya ikan air tawar di Provinsi Banten dan mengevaluasi kepekaan E. coli terhadap tiga jenis antibiotik, yaitu tetrasiklin, oksitetrasiklin, dan enrofloksasin. Isolat E. coli diidentifikasi se- cara uji biokimia menggunakan vitek 2 compact, kemudian uji kepekaan E. coli terhadap tiga jenis antibiotik dilakukan secara in vitro menggunakan metode difu- si (uji cakram). Isolat E. coli yang resisten terhadap antibiotik tertentu dilanjutkan dengan uji konsentrasi hambat minimum (KHM). Hasil uji di interpretasi menurut Clinical Laboratory Standards Institute (CLSI). Berdasarkan hasil penelitian dipe- roleh 45 isolat E. coli dengan persentase tingkat resisten uji difusi terhadap tetra- siklin sebesar 33,33%, oksitetrasiklin sebesar 35,56% dan enrofloksasin sebesar 15,55%. Diketahui 16 isolat E. coli mengalami resisten. Lima belas isolat resisten terhadap tetrasiklin memiliki nilai KHM yang berbeda, di antaranya 2 isolat me- miliki nilai KHM >256 μg/mL, 11 isolat memiliki nilai KHM 256 μg/mL dan 2 isolat memiliki nilai KHM 128 μg/mL. Enam belas isolat mengalami resisten ter- hadap oksitetrasiklin memiliki nilai KHM yang berbeda, di antaranya 10 isolat memiliki nilai KHM >256 μg/mL, 5 isolat memiliki nilai KHM 256 μg/mL dan 1 isolat memiliki nilai KHM 128 μg/mL. Tujuh isolat mengalami resisten terhadap enrofloksasin memiliki nilai KHM berbeda, di antaranya 3 isolat memiliki nilai KHM 128 μg/mL, 3 isolat memiliki nilai KHM 64 μg/mL dan 1 isolat memiliki nilai KHM 16 μg/mL.
