Abstrak

Penelitian ini bertujuan untuk mengisolasi senyawa flavonoid yang bersifat sebagai insektisida nabati dari ekstrak metanol daun gamal (Gliricidia maculata, L.). Ekstrak metanol diperoleh dengan cara maserasi serbuk daun gamal. Fraksinasi dan pemurnian ekstrak metanol dilakukan dengan metode kromatografi kolom dengan adsorben Sephadex LH-20 dan eluen MeOH : H2O (4:1). Fraksi– fraksi yang dihasilkan dipantau menggunakan metode kromatografi lapis tipis dengan adsorben Silica Gel 60 F254 pada plat alumunium dan eluen DCM : MeOH (7:3) dan diidentifikasi dengan larutan AlCl3 pada Rf 0,531. Fraksi tersebut direkristalisasi dan diperoleh kristal RIO-46A berbentuk amorf dan berwarna putih kekuningan dengan berat 0,35 gram. Analisis kristal menggunakan spektrofotometer UV-Vis menunjukkan adanya serapan maksimum pada λ 268 (pita II) dan λ 349 (pita I) dan analisis menggunakan spektrofotometer IR memberikan puncak absorbsi pada bilangan gelombang 3373 cm-1 (O-H), 2926 - 2885 cm-1 (C-H alifatik), 1593 - 1459 cm -1 (C=C aromatik), 1729 cm-1 (C=O) dan 1073 cm-1 (C-O-C). Berdasarkan kedua metode spektrofotometri tersebut, dapat disimpulkan bahwa isolat merupakan aglikon flavonoid dari golongan flavon. Hasil dari uji bioassay, aglikon flavonoid dari golongan flavon ini memiliki aktivitas biologis sebagai insektisida nabati terhadap hama kutu putih tanaman pepaya (Paracoccus marginatus) dengan nilai LC50 3,35% dalam waktu 12 jam setelah perlakuan. Kata kunci : senyawa flavonoid, isolasi, daun gamal, insektisida nabati (This reseach aims to isolate flavonoid compound which has botanical insecticide from metanol extract of Gliricidia maculata leaves. The methanol extract was obtained from maceration of G. maculate leaves powder. Fractination and purification of the extract was done with column chromatography method using Sephadex LH-20 as adsorbent and MeOH : H2O (4:1) as eluent. The fractions resulted in was monitored by thin layer chromatography method with adsorbent of Silica Gel 60 F254 on alumunium plate and DCM : MeOH (7:3) as eluent. Identification of flavonoid compound using AlCl3 gave Rf value of 0,531. The relatively pure fraction was recrystallized and gave yellowish white crystal RIO-46A about 0,35 gram. Analysis of the crystal using UV-Vis spectrophotometry showed maximum adsorption at λ 268 (band II) and λ 349 (band I) and analysis using IR spectrophotometry gave adsorption bands on wavelength numbers of 3373 cm-1 (O-H), 2926-2885 cm-1 (aliphatic C-H), 1593-1459 cm-1 (C = C aromatic), 1729 cm-1 (C = O) and 1073 cm-1 (C-O-C). Based on both analysis method, it is concluded that isolate was a flavonoid aglycon from flavon. The result of bioassay test toward of papaya mealybug (Paracoccus marginatus) showed that the isolate has a characteristic of botanical insecticide with LC50 3,35% after 12 hours of treatment.
