Abstract
Background: Cashew leaves (Anacardium occidentale L.) possess antibacterial, antioxidant, and anti-inflammatory activities. However, the development of cashew leaf extract is limited by the poor aqueous solubility of its bioactive compounds. Therefore, one approach to enhance bioavailability is the formulation of a Self Nano Emulsifying Drug Delivery System (SNEDDS). Objective: This study aimed to optimize the SNEDDS formula based on 96% ethanol extract of cashew leaves with varying concentrations of Smix (Tween 80 and PEG 400) using the Simplex Lattice Design (SLD) method. Methods: The experimental study used Design-Expert version 13. Evaluation parameters included % transmittance, emulsification time, pH, and viscosity. The optimal formula was determined based on desirability values, followed by verification and characterization tests for particle size, polydispersity index (PDI), and zeta potential. Result: Variations in Smix in the initial formula significantly affected all evaluation parameters (p<0.05). The optimal formula had a desirability value of 0.876 with a transmittance of >98%, an emulsification time of <1 minute, a pH of ±5.05 to ±5.22, and a viscosity of ±72 to ±76 cPs. Verification tests showed no significant difference between the predicted and actual results (p>0.05). Characterization showed a particle size of 67 nm, a PDI of 0.191, and a zeta potential of -32,56 mV, and the ability to form a stable SNEDDS system. Conclusion : The SLD method is effective in determining the optimal SNEDDS formula with physical characteristics and stability that meet the criteria for a nanoemulsion. Latar Belakang: Daun jambu mete (Anacardium occidentale L.) memiliki aktivitas antibakteri, antioksidan, dan antiinflamasi, namun pemanfaatannya masih terbatas akibat kelarutan senyawa bioaktif yang rendah. Salah satu strategi peningkatan bioavailabilitas adalah melalui formulasi Self Nano Emulsifying Drug Delivery System (SNEDDS). Tujuan: Penelitian ini bertujuan mengoptimasi formula SNEDDS berbasis ekstrak etanol 96% daun jambu mete dengan variasi konsentrasi Smix (Tween 80 dan PEG 400) menggunakan metode Simplex Lattice Design (SLD). Metode: Penelitian eksperimental menggunakan Design-Expert versi 13. Parameter evaluasi meliputi % transmitansi, waktu emulsifikasi, pH, dan viskositas. Formula optimal ditentukan berdasarkan nilai desirabilitas, dilanjutkan uji verifikasi dan karakterisasi ukuran partikel, indeks polidispersitas (PDI), dan potensial zeta. Hasil: Variasi Smix pada formula awal berpengaruh signifikan terhadap seluruh parameter evaluasi (p<0,05). Formula optimal memiliki nilai desirabilitas 0,876 dengan %transmitansi >98%, waktu emulsifikasi <1 menit, pH ±5,05 sampai ±5,22, dan viskositas ±72 sampai ±76 cPs. Uji verifikasi menunjukkan tidak terdapat perbedaan signifikan antara prediksi dan hasil aktual (p>0,05). Karakterisasi menunjukkan ukuran partikel 67 nm, PDI 0,191 dan potensial zeta -32,56 mV serta mampu membentuk sistem SNEDDS yang stabil. Simpulan: Metode SLD efektif dalam menentukan formula SNEDDS optimal dengan karakteristik fisik dan stabilitas yang memenuhi kriteria nanoemulsi.
