N2  - Dalam penelitian ini telah dilakukan isolasi dan uji aktivitas antioksidan ekstrak
metanol dari batang binahong (Anredera cordifolia). Proses isolasi meliputi
tahapan maserasi, analisis HPLC dan analisis MPLC dengan menggunakan eluen
metanol : air (7:3). Hasil MPLC menghasilkan 19 fraksi dimana puncak senyawa
terdapat pada fraksi 4,5 dan 6. Senyawa hasil isolasi merupakan suatu kristal
amorf dari fraksi 5 sebanyak 10 mg. Penentuan struktur senyawa dilakukan
dengan spektrofotometri UV-Vis dengan penambahan pereaksi geser NaOH.
Hasil analisis senyawa mengidentifikasi suatu golongan flavonoid yang memiliki
kerangka senyawa flavon. Pada uji aktivitas antioksidan dengan metode
voltammetri siklik bahwa isolat RNV-1 menghasilkan nilai koefisien aktivitas
antioksidan (slope) lebih besar dibandingkan dengan kontrol positif asam askorbat
yaitu sebesar 0,055 (isolat RNV-1) dan 0,052 (asam askorbat).

ABSTRACT

In this study has been conducted isolation and determination of antioxidant
activity of methanol extract of the stem binahong (Anredera cordifolia). The
isolation process includes stages of maceration, analysis HPLC and analysis
MPLC by using eluent methanol : water (7: 3). The MPLC Results produce 19
fractions where the peak of compounds contained in fractions 4,5 and 6. The
isolated Compounds a amorf crystal from fraction 5 as much as 10 mg. The
structural determination is done by UV-Vis spectrophotometry with the addition
of NaOH sliding reagent. The results of analysis of compounds to identify a class
of flavonoids that have the skeleton of the flavon. The antioxidant activity test
with cyclic voltammetry method that RNV-1 isolates produced antioxidant
activity coefficient greater than the positive control ascorbic acid that is equal to
0.055 (RNV-1 crystal isolates) and 0.052 (ascorbic acid).
