"This research aims to study the effect of addition sorbitol toward stability cellulose\r\nenzyme from Rhizopus oryzae. To approach this aims, the enzyme has been done\r\nto produce, to isolate and to purify. The purification of cellulose enzyme was\r\ndone by fractination using the ammonium sulfat and dialysis. The purified enzyme\r\nwas mixed by sorbitol. Determination of the cellulose enzyme activity performed\r\nby the method of Mandels, while the measurement of protein concentration\r\nperformed by the method of Lowry.\r\nThe results showed that the purified enzyme has specific activity 9.18 U/mg,\r\nincreased 4.03 times than the native enzyme. The test of thermal stability of the\r\nenzyme at temperature 50°C for 60 minutes has k\r\ni\r\n= 0.02 min\r\n-1\r\n; t\r\n= 34.65 min\r\nand ΔG\r\ni\r\n= 99.7 kJ mol\r\n-1\r\n. The test of thermal stability of the enzyme mixed by\r\nsorbitol 0.5; 1.0; and 1.5 M at temperature 50°C for 60 minutes each has t\r\n= 49.5\r\nmin, k\r\ni\r\n= 0.014 min\r\n-1\r\nand ΔG\r\ni\r\n= 100. kJ mol\r\n;\r\nt\r\n1/2\r\n= 24.75 min, k\r\ni\r\n= 0.028 min\r\n-1\r\nand ΔG\r\ni\r\n-1\r\n= 101.8 kJ mol\r\n-1\r\n; t\r\n= 22.35 min,\r\nki = 0.031 min\r\n-1\r\nand ΔGi = 100.8 kJ mol\r\nThe results showed that the purified enzyme has K\r\n-1\r\n.\r\nm\r\nand V\r\nvalues\r\n74.02 mg/mL substrate and 6.57\r\n1/2\r\nmax\r\n1/2\r\nμmol/mL.min\r\n, and the enzyme mixed by sorbitol\r\n0.5; 1.0; and 1.5 M each has\r\n24.89 mg/mL substrate and 0.92 μmol/mL.min;\r\n18.39 mg/mL substrate and 0.82 μmol/mL.min; 14.83 mg/mL substrate and 0.42\r\nμmol/mL.min.\r\nAlthough the optimum of pH and temperature of the mixed enzyme by sorbitol did\r\nnot changed, at pH 7 and at temperature 60\r\no\r\nC but the thermal stability of\r\nmodified enzyme was increased and showed by the decrease in the value of k\r\ni\r\nin accordance with the increase half life and ΔG\r\nKey word: Cellulase enzyme, sorbitol, Rhizopus oryzae\r\ni\r\n.\r\n1/2\r\n\r\n\r\nABSTRAK\r\n\r\n\r\nPenelitian ini bertujuan untuk mempelajari pengaruh penambahan sorbitol terhadap\r\nstabilitas enzim selulase dari Rhizopus oryzae. Untuk mencapai tujuan tersebut\r\ndilakukan produksi, isolasi, dan pemurnian enzim. Pemurnian enzim selulase dilakukan\r\ndengan menggunakan fraksinasi dengan ammonium sulfat dan dialisis. Enzim hasil\r\npemurnian kemudian ditambahkan sorbitol dengan konsentrasi 0,5 M; 1,0 M; dan 1,5 M.\r\nPenentuan aktivitas enzim selulase dilakukan dengan metode Mandels sedangkan\r\npengukuran kadar protein dilakukan dengan metode Lowry.\r\nHasil penelitian menunjukkan bahwa enzim hasil pemurnian memiliki aktivitas spesifik\r\nsebesar 9,18 U/mg, meningkat 4,03 kali dibandingkan ekstrak kasar enzim. Uji stabilitas\r\ntermal enzim hasil pemurnian pada suhu 50\r\n;t\r\n1/2\r\n= 34,65 menit; dan ∆G\r\ni\r\no\r\nC selama 60 menit memiliki k\r\n= 99,7 kJmol\r\n-1\r\n. Uji stabilitas termal enzim setelah\r\npenambahan sorbitol 0,5; 1,0; dan 1,5 M pada suhu 50\r\no\r\nC selama 60 menit memiliki\r\nberturut-turut t\r\n1/2\r\n= 49,5 menit, k\r\ni\r\n= 0,014 menit\r\n-1\r\ndan\r\n∆G\r\n= 24,75\r\nmenit, k\r\ndan\r\n∆G\r\ni\r\ni\r\n= 0,028 menit\r\n= 100,8 kJmol\r\n-1\r\n-1\r\ndan\r\n∆\r\nG\r\ni\r\n= 101,8 kJmol\r\n-1\r\n;\r\nt\r\n1/2\r\ni\r\n= 100 kJmol\r\n= 22,35 menit, k\r\n.\r\nEnzim hasil pemurnian memiliki nilai Km dan Vmaks berturut-turut sebesar 74,02\r\nmg/mL substrat dan 6,57 μmol/mL.menit, sedangkan enzim setelah peambahan sorbitol\r\n0,5; 1,0; dan 1,5 M\r\nmemiliki nilai Km dan Vmaks berturut-turut sebesar 24,89 mg/mL\r\nsubstrat dan 0,92 μmol/mL.menit; 18,39 mg/mL substrat dan 0,82 μmol/mL.menit;\r\n14,83 mg/mL substrat dan 0,42 μmol/mL.menit.\r\nWalaupun suhu optimum dan pH optimum enzim setelah penambahan sorbitol tidak\r\nmengalami perubahan, yakni tetap pada pH 7 dan suhu 60\r\no\r\nC tetapi terjadi peningkatan\r\nstabilitas termal karena adanya penurunan nilai k\r\n∆G\r\ni\r\n.\r\nKata Kunci: Enzim selulase, sorbitol, Rhizopus oryzae.\r\ni\r\ni\r\n= 0,02 menit\r\n-1\r\n; t\r\n1/2\r\n= 0,031 menit\r\ni\r\nserta peningkatan waktu paruh dan\r\n-1\r\n-1"^^ .
