?url_ver=Z39.88-2004&rft_id=1517011006&rft_val_fmt=info%3Aofi%2Ffmt%3Akev%3Amtx%3Adc&rft.title=PRODUKSI+BIOSURFAKTAN+DARI+BAKTERI+ISOLAT+LOKAL+BSPP-4+ASAL+SEDIMEN+PERAIRAN+PELABUHAN+PANJANG+DAN+UJI+POTENSI+ANTIBAKTERI&rft.creator=WIWIN+INDRIANTI%2C+1517011006&rft.subject=001+Ilmu+pengetahuan&rft.description=Biosurfaktan+adalah+produk+metabolit+sekunder+yang+dihasilkan+oleh+mikroba.+Biosurfaktan+banyak+diaplikasikan+dalam+berbagai+bidang%2C+salah+satunya+dalam+bidang+medis+yang+biasa+digunakan+sebagai+antibakteri%2C+antifungi%2C+dan+antivirus.+Penelitian+ini+bertujuan+untuk+mendapatkan+biosurfaktan+dari+bakteri+isolat+lokal+BSPP-4+asal+sedimen+perairan+Pelabuhan+Panjang%2C+serta+menguji+potensinya+sebagai+antibakteri.+Isolat+lokal+BSPP-4+diremajakan+menggunakan+nutrient+agar.+Metode+uji+yang+dilakukan+meliputi+uji+biosurfaktan+(hemolisis%2C+IE24%2C+Oil+Spread%3DOS%2C+dan+Drop+Collapse%3DDC)%2C+karakterisasi+biokimia+bakteri+isolat+BSPP-4%2C+pembuatan+kurva+pertumbuhan%2C+optimasi+produksi+biosurfaktan%2C+produksi+biosurfaktan%2C+karakterisasi+biosurfaktan+menggunakan+FT-IR%2C+serta+uji+potensi+biosurfaktan+sebagai+antibakteri.+Hasil+uji+biosurfaktan+diketahui+bahwa+isolat+BSPP-4+memiliki+aktivitas+%EF%81%A2-hemolisis%2C+dengan+nilai+IE24+33%25%2C+uji+OS+2+mm%2C+dan+uji+DC+positif.+Karakterisasi+biokimia+menunjukkan+bahwa+isolat+BSPP-4+merupakan+jenis+bakteri+Bacillus.+Optimasi+produksi+menunjukkan+kondisi+optimum+untuk+produksi+biosurfaktan+pada+waktu+inkubasi+96+jam%2C+sumber+karbon+gliserol+2%25%2C+sumber+nitrogen+NaNO3+0%2C3%25%2C+dan+konsentrasi+inokulum+7%2C5%25.+Produksi+biosurfaktan+dari+bakteri+isolat+BSPP-4+memilki+ciri-ciri+fisik+biosurfaktan+berwarna+kuning+tua+sebanyak+0%2C9674+g%2FL.+Hasil+Analisis+data+FT-IR+memperlihatkan+adanya+serapan+dari+gugus+fungsi%2C+seperti+pada+daerah+3272+cm-1+(N-H+Stretching)%2C+1535+cm-1+(deformasi+ikatan+N-H)%2C+2937+cm-1+(C-H+stretching)%2C+1625+cm-1+(CO-N+stretching)%2C+pada+daerah+1103+cm-1+dan+1185+cm-1+(C-O+Stretching)%2C+daerah+2937+cm-1%2C+dan+1625+cm-1+(rantai+alifatik+C-H)+yang+merupakan+ciri-ciri+surfaktan+lipopeptida.+Uji+antibakteri+ekstrak+biosurfaktan+menunjukkan+adanya+zona+bening+sebesar+1%2C72+cm+pada+bakteri+Gram+negatif+Escherichia+coli%2C+sedangkan+pada+bakteri+Gram+positif+Staphylococcus+aureus+sebesar+1%2C16+cm.+Berdasarkan+hasil+penelitian+ini%2C+dapat+disimpulkan+bahwa+isolat+BSPP-4+diduga+menghasilkan+biosurfaktan+jenis+lipopeptida+dan+memilliki+kemampuan+sebagai+antibakteri.%0D%0A%0D%0AKata+Kunci+%3A+Biosurfaktan%2C+lipopeptida%2C+antibakteri%2C+FT-IR%2C+Indeks+Emulsi.%0D%0A%0D%0ABiosurfactants+are+secondary+metabolite+products+from+microbes.+Biosurfactants+are+widely+applied+in+various+fields%2C+one+of+which+is+in+the+medical+field+which+is+commonly+used+as+antibacterial%2C+antifungal%2C+and+antivirus.+This+study+aims+to+obtain+biosurfactants+from+local+isolates+of+bacteria+BSPP-4+from+the+sediments+of+Panjang+Port+waters%2C+and+to+test+their+potential+as+antibacterial.+Local+isolates+of+BSPP-4+bacteria+was+maintenances+in+nutrient+agar+medium.+The+analysis+include+biosurfactant+tests+(hemolysis%2C+IE24%2C+Oil+Spread%3DOS%2C+and+Drop+Collapse%3DDC)%2C+biochemical+characterization+of+BSPP-4+isolate+bacteria%2C+growth+curve+creation%2C+optimization+of+biosurfactant+production%2C+production+of+biosurfactant%2C+characterization+of+biosurfactants+using+FT-IR%2C+as+well+as+testing+the+potential+of+biosurfactants+as+antibacterial.+The+biosurfactant+test+showed+that+the+BSPP-4+isolate+had+%EF%81%A2-hemolysis%2C+with+an+IE24+value+of+33%25%2C+2+mm+OS+test%2C+and+a+positive+DC+test.+Biochemical+characterization+showed+that+BSPP-4+isolate+was+a+type+of+Bacillus+bacteria.+The+optimum+conditions+for+biosurfactant+production+was+at+an+incubation+time+of+96+hours%2C+2%25+glycerol+carbon+source%2C+0.3%25+NaNO3+nitrogen+source%2C+and+7.5%25+inoculum+concentration.+The+production+of+biosurfactant+of+bacterial+isolate+BSPP-4+has+the+physical+characteristics+of+a+dark+yellow+biosurfactant+as+much+as+0.9674+g%2FL.+The+FT-IR+analysis+data+showed+that+there+was+absorption+from+functional+groups%2C+such+as+in+the+area+of+3272+cm-1+(NH+Stretching)%2C+1535+cm-1+(NH+bond+deformation)%2C+2937+cm-1+(CH+stretching)%2C+1625+cm-1+(CO-N+stretching)%2C+on+the+area+1103+cm-1+and+1185+cm-1+(CO+Stretching)%2C+2937+cm-1+area+and+1625+cm-1+(CH+aliphatic+chain)+which+are+characteristics+of+lipopeptide+surfactants.+Antibacterial+test+of+biosurfactant+extract+showed+a+clear+zone+of+1.72+cm+on+Gram+negative+bacteria+Escherichia+coli%2C+while+in+Gram+positive+bacteria+Staphylococcus+aureus+of+1.16+cm.+Based+on+the+results+of+this+study%2C+it+can+be+concluded+that+BSPP-4+isolate+can+produce+lipopeptide+biosurfactant+and+has+the+ability+as+an+antibacterial.%0D%0AKeywords%3A+Biosurfactant%2C+lipopeptide%2C+antibacterial%2C+FT-IR%2C+Emulsification+indeks%0D%0A&rft.publisher=FAKULTAS+MATEMATIKA+DAN+ILMU+PENGETAHUAN+ALAM&rft.date=2022-02-11&rft.type=Skripsi&rft.type=NonPeerReviewed&rft.
