"Tanaman sawi caisim (Brassica rapa L.) sudah dikenal luas di Indonesia.\r\nPengembangan budidaya sawi memiliki prospek baik untuk mendukung upaya\r\npeningkatan pendapatan petani, peningkatan gizi masyarakat, dan pengembangan\r\nagribisnis secara luas. Salah satu cara untuk mengembangkan budidaya sawi\r\ncaisim yaitu dengan menguji tanaman yang toleran atau resisten terhadap berbagai\r\ncekaman dari lingkungan, dalam hal ini cekaman garam. Tujuan penelitian ini\r\nadalah mengetahui konsentrasi NaCl yang efektif untuk seleksi planlet sawi\r\ncaisim dalam kondisi cekaman garam secara in vitro, serta mengetahui karakter\r\nekspresinya. Penelitian ini dilaksanakan dari bulan Februari 2021 sampai dengan\r\nApril 2021 di ruang Kultur In Vitro, Laboratorium Botani, Jurusan Biologi,\r\nFakultas Matematika dan Ilmu Pengetahuan Alam, Universitas Lampung.\r\nRancangan penelitian ini disusun menggunakan Rancangan Acak Lengkap (RAL)\r\nyang terdiri dari faktor tunggal yaitu konsentrasi NaCl dengan 5 taraf perlakuan:\r\n0%; 0,25%; 0,50%; 0,75%; dan 1% dengan 5 kali ulangan. Analisis data\r\nmenggunakan uji Homogenitas Ragam dan uji ANOVA, serta uji lanjut\r\nmenggunakan uji Tukey pada taraf nyata 5%. Hasil penelitian menunjukkan\r\nbahwa konsentrasi NaCl yang efektif dalam mensimulasikan kondisi cekaman\r\ngaram untuk seleksi planlet sawi caisim secara in vitro adalah 0,50%. Hasil\r\nkarakterisasi planlet sawi caisim pada kondisi cekaman garam menunjukkan\r\nbahwa semakin tinggi konsentrasi NaCl, maka ukuran planlet semakin kecil\r\ndengan tinggi planlet yang semakin rendah, visualisasi warna daun menjadi hijau\r\nkekuningan, akar planlet yang semakin panjang, serta kandungan klorofil a, b, dan\r\nklorofil total yang semakin menurun.\r\nKata Kunci: Cekaman garam, in vitro, NaCl, Brassica rapa\r\nField mustard plant (Brassica rapa L.) is well known in Indonesia. The\r\ndevelopment of mustard cultivation has good prospects to support efforts to\r\nincrease farmers income, improve community nutrition, and develop agribusiness\r\nin general. One of the ways to develop field mustard cultivation is to test plants\r\nthat are tolerant or resistant to various stresses from the environment, in this case\r\nsalinity stress. The purpose of this study was to determine the effective\r\nconcentration of NaCl for the selection of field mustard plantlets under salinity\r\nstress conditions in vitro, as well as to determine the character of its expression.\r\nThis research was conducted from February 2021 to April 2021 in the In Vitro\r\nCulture Room, Botany Laboratory, Department of Biology, Faculty of\r\nMathematics and Natural Sciences, University of Lampung. The research design\r\nwas prepared using a completely randomized design (CRD) consisting of a single\r\nfactor, which is concentration of NaCl with 5 levels of treatment: 0%; 0,25%;\r\n0,50%; 0,75%; and 1% with 5 repetitions. Data analysis used the homogeneity test\r\nof variance and ANOVA test, and further tests used the Tukey test at the 5% real\r\nlevel. The results showed that the effective concentration of NaCl for the selection\r\nof field mustard plantlets under salinity stress conditions was 0,50%. The results\r\nof the characterization of field mustard plantlets in salinity stress conditions\r\nshowed that the higher the NaCl concentration, then the smaller the plantlet size,\r\nthe lower the plantlet height, the smaller the leaf size with the visualization of the\r\nleaf color becoming yellowish green, the longer plantlet roots, and the decreasing\r\ncontent of chlorophyll a, b and total chlorophyll.\r\n\r\nKeywords: salt stress, in vitro, NaCl, Brassica rapa"^^ .
