Namun, serat kasar lignoselulosa pada ampas tahu dapat mengurangi efisiensi degradasi oleh enzim hidrolisis. Perlakuan awal (pretreatment) menggunakan ragi oncom dilakukan untuk membantu proses hidrolisis serat kasar. Ragi oncom mengandung kapang Neurospora sitophila yang mampu menghasilkan enzim lignoselulotik untuk membantu menguraikan senyawa lignoselulosa. Penelitian ini bertujuan untuk mengetahui peningkatan biodegradability ampas tahu dengan pemberian ragi oncom. Penelitian yang dilakukan menggunakan metode deskriptif dengan dua faktor yaitu konsentrasi ragi (0%, 2%, 4%, 6%) dan lama waktu pretreatment (0 jam, 24 jam, 48 jam, 72 jam, 96 jam). Pengamatan yang dilakukan meliputi derajat keasaman (pH), Total Solid (TS), Total Volatile Acid (TVA), Soluble Chemical Oxygen Demand (S-COD), dan konsentrasi lignoselulosa. Hasil penelitian yang diperoleh adalah terjadinya peningkaan biodegradability dengan meningkatnya nilai S-COD mencapai 28.098 mg/L dan nilai TVA sebesar 8.840 mg/L. Selain itu, terjadi penurunan kondisi pH mencapai 5,25, nilai TS sebesar 8,03%, konsentrasi selulosa, hemiselulosa, dan lignin berturut-turut sebesar 14,51%, 35,42%, dan 7,73%. However, lignocellulosic crude fiber in tofu dreg can reduce the efficiency of degradation by hydrolytic enzymes. Initial treatment (pretreatment) using oncom yeast was carried out to help the hydrolysis process of crude fiber. Oncom yeast contains the mold Neurospora sitophila which is capable of producing lignocellulotic enzymes to help decompose lignocellulosic compounds. This study aims to improve the biodegradability of tofu dreg by administering oncom yeast. The research was conducted using a descriptive method with two factors, namely yeast concentration (0%, 2%, 4%, 6%) and length of pretreatment time (0 hour, 24 hours, 48 hours, 72 hours, 96 hours). Observations made included the degree of acidity (pH), Total Solid (TS), Total Volatile Acid (TVA), Soluble Chemical Oxygen Demand (S-COD), and concentration of lignocellulose. The research results obtained were an increase in biodegradability with an increase in the S-COD value reaching 28.098 mg/L and the TVA value of 8.840 mg/L. In addition, there was a decrease in pH conditions reaching 5,25, TS value of 8,03%, concentrations of cellulose, hemicellulose, and lignin respectively 14,51%, 35,42%, and 7,73%.
