Bakteri Bacillus thuringiensis (Bt) memiliki\r\nkarakteristik yang berbeda dengan spesies Bacillus lain, yaitu dapat menghasilkan\r\nkristal protein yang bersifat toksik bagi serangga pada fase sporulasi. Kristal\r\ninsektisida tersebut terdiri dari satu atau lebih protein kristal (Cry) dan sitolitik\r\n(Cyt), yang dikenal sebagai δ-endotoksin. Spesifisitas dan aktivitas Bacillus\r\nthuringiensis terhadap inangnya dikaitkan dengan jenis dan bentuk δ-endotoksin\r\nyang dihasilkan. Penelitian ini dilakukan untuk mengetahui awal terbentuknya\r\nkristal protein pada waktu pertumbuhan bakteri serta karakter bentuk kristal protein\r\ndari enam isolat Bacillus thuringiensis yang diisolasi dari tanah Kebun Raya Liwa\r\n(KRL) Lampung. Metode penelitian yang digunakan adalah deskriptif kualitatif\r\nyang dipresentasikan dalam bentuk gambar dan tabel. Identifikasi morfologi kristal\r\nprotein bakteri Bacillus thuringiensis selama fase sporulasi dilakukan dengan\r\nmenggunakan Scanning Electron Microscope (SEM). Sedangkan pengamatan\r\nwaktu terbentuknya kristal dilakukan dengan metode pengecatan menggunakan\r\nComassie Brilliant Blue dan Crystal Violet dengan interval waktu setiap tiga jam\r\nsekali selama 48 jam. Gambar hasil SEM menunjukkan bahwa setiap isolat\r\nmemiliki lebih dari satu bentuk kristal protein dengan tujuh variasi morfologi kristal\r\nprotein, yaitu ovoid (oval), spherical (bulat), bipiramidal segitiga, rectangle (empat\r\npersegi panjang), elongate (memanjang), cuboid (kotak), serta geometrical.\r\nTerbentuknya kristal protein pada keenam isolat menunjukkan waktu yang berbeda.\r\nSecara keseluruhan kristal protein sudah terlihat pada jam pertumbuhan ke-3 hingga\r\nke-18. Waktu terbentuknya kristal tersebut dapat dijadikan sebagai referensi untuk\r\npengujian aktivitas toksisitas isolat.\r\nKata kunci : Bacillus thuringiensis, kristal protein, morfologi, Scanning Electron\r\nMicroscope, waktu terbentuknya kristal\r\n\r\nBacillus is a Gram-positive rod-shaped bacterium and is often used in the field of\r\nbiotechnology. Bacillus thuringiensis (Bt) bacteria can produce protein crystals that\r\nare toxic to insects during the sporulation phase. The insecticide crystals consist of\r\none or more crystalline (Cry) and cytolytic (Cyt) proteins, which are known as δ-\r\nendotoxins. The specificity and activity of Bacillus thuringiensis on the host are\r\nrelated to the type and form of the δ-endotoxin produced. This research was\r\nconducted to determine the initial formation of protein crystals during bacterial\r\ngrowth and the character of the protein crystal shape of six isolates of Bacillus\r\nthuringiensis isolated from the soil of the Liwa Botanical Garden Lampung. The\r\nresearch method used is descriptive qualitative which is presented in the form of\r\npictures and tables. Identification of the protein crystal morphology of Bacillus\r\nthuringiensis was carried out using a Scanning Electron Microscope (SEM). While\r\nobserving the time of crystal formation was carried out by staining method using\r\nComassie Brilliant Blue and Crystal Violet with intervals every three hours for 48\r\nhours. SEM results show that each isolate has more than one protein crystal shape\r\nwith seven variations of protein crystal morphology, namely ovoid, spherical,\r\ntriangular bipyramidal, rectangular, elongate, cuboid, as well as geometrical. The\r\nformation of protein crystals in the six isolates showed different times. The time of\r\nprotein crystals formation were seen at the 3rd to 18th hour of growth.
