?url_ver=Z39.88-2004&rft_val_fmt=info%3Aofi%2Ffmt%3Akev%3Amtx%3Adc&rft.title=IMOBILISASI+ENZIM+LIPASE+DARI+ISOLAT+BAKTERI+Klebsiella+sp.%0D%0ALPG172+SECARA+ADSORPSI+MENGGUNAKAN+MATRIKS+ZEOLIT%0D%0A%0D%0AALAM+LAMPUNG&rft.creator=Fitriana+%2C+Artika+Sari&rft.subject=500+ilmu+pengetahuan+alam+dan+matematika&rft.subject=540+Kimia+dan+ilmu+yang+berhubungan&rft.description=Dalam+penelitian+ini%2C+dilakukan+imobilisasi+enzim+lipase+dari+bakteri%0D%0AKlebsiella+sp.+LPG172+menggunakan+matriks+zeolit+alam+Lampung.+Tahap%0D%0Apenelitian+dilakukan+dengan+memproduksi+ekstrak+kasar+lipase+yang+kemudian%0D%0Adimurnikan+dengan+fraksinasi+ammonium+sulfat%2C+dialisis+dan+kromatografi+kolom%0D%0Afiltrasi+gel+sephadex+G-75.+Lipase+hasil+pemurnian+diimobilisasi+menggunakan%0D%0Azeolit+alam+Lampung+teraktivasi+dan+ditentukan+kondisi+optimumnya.+Ekstrak+kasar%0D%0Aenzim+lipase+mempunyai+aktivitas+sebesar+34%2C60+U%2FmL+dengan+kadar+protein%0D%0Asebesar+3%2C34+mg%2FmL.+Fraksi+enzim+hasil+pemurnian+dengan+kromatografi+kolom%0D%0Afiltrasi+gel+sephadex+G-75+memiliki+kadar+protein+sebesar+0%2C69+mg%2FmL+dengan+nilai%0D%0Aaktivitas+hidrolisis+serta+transesterifikasi+masing-masing+sebesar+1.615%2C33+U%2FmL%0D%0Adan+1.178%2C80+U%2FmL.+Setelah+diimobilisasi+menggunakan+matriks+zeolit+alam%0D%0A%0D%0ALampung+teraktivasi+aktivitas+hidrolisis+dan+transesterifikasi+enzim+lipase+masing-%0D%0Amasing+menjadi+1.205%2C67+U%2FmL+dan+1.108%2C80+U%2FmL.%0D%0A%0D%0AProses+imobilisasi+lipase+Klebsiella+sp.+LPG172+menggunakan+zeolit+alam%0D%0ALampung+teraktivasi+mengubah+kondisi+optimum+pada+kedua+aktivitas+enzim.+Pada%0D%0Areaksi+hidrolisis%2C+lipase+bebas+memiliki+kondisi+optimum+pH+7%2C+suhu+80+%CC%8AC+dengan%0D%0Awaktu+inkubasi+selama+25+menit.+Namun%2C+setelah+diimobilisasi+lipase+bekerja+secara%0D%0Aoptimal+pada+pH+8%2C+suhu+50+%CC%8AC+serta+waktu+inkubasi+selama+15+menit.+Sedangkan%0D%0Adalam+reaksi+transesterifikasi%2C+lipase+bebas+memiliki+kondisi+optimum+pada+pH+7%2C%0D%0Asuhu+70+%CC%8AC+dengan+waktu+inkubasi+25+menit.+Setelah+diimobilisasi+lipase+bekerja%0D%0Asecara+optimal+pada+pH+8%2C+suhu+60+%CC%8AC+dengan+waktu+inkubasi+selama+10+menit.%0D%0AKondisi+optimum+yang+diperoleh+digunakan+dalam+uji+stabilitas+lipase+imobil%0D%0Aterhadap+pemakaian+berulang.+Pada+reaksi+hidrolisis%2C+lipase+imobil+dapat+dilakukan%0D%0Apemakaian+2+kali+dalam+siklus+reaksi+dengan+aktivitas+sisa+sebesar+43%2C09%25+dari%0D%0Aaktivitas+awal.+Sedangkan+pada+reaksi+transesterifikasi+pemakaian+enzim+lipase%0D%0Aimobil+dapat+dilakukan+sebanyak+3+kali+siklus+reaksi+dengan+persen+aktivitas+akhir%0D%0Asebesar+55%2C98%25.%0D%0A%0D%0AKata+kunci%3A+hidrolisis%2C+imobilisasi%2C+Klebsiella+sp.+LPG172%2C+lipase%2C+transesterifikasi%2C%0D%0A%0D%0Azeolit+alam+Lampung.%0D%0AIn+this+research%2C+the+immobilization+of+lipase+from+Klebsiella+sp.+LPG172+was%0D%0Acarried+out+using+natural+zeolite+from+Lampung.+The+research+involved+producing+a%0D%0Acrude+lipase+extract%2C+which+was+subsequently+purified+through+ammonium+sulfate%0D%0Afractionation%2C+dialysis%2C+and+sephadex+G-75+gel+filtration+chromatography.+The%0D%0Apurified+lipase+was+then+immobilized+using+activated+natural+zeolite+from+Lampung%2C%0D%0Aand+its+optimal+conditions+were+determined.+The+crude+lipase+extract+exhibited+an%0D%0Aactivity+of+34.60+U%2FmL+with+a+protein+concentration+of+3.34+mg%2FmL.+The+enzyme%0D%0Afraction+obtained+from+sephadex+G-75+gel+filtration+chromatography+had+a+protein%0D%0Aconcentration+of+0.69+mg%2FmL%2C+with+hydrolysis+and+transesterification+activities+of%0D%0A1615.33+U%2FmL+and+1178.80+U%2FmL%2C+respectively.+After+immobilization+with%0D%0Aactivated+natural+zeolite+from+Lampung%2C+the+lipase+hydrolysis+and%0D%0Atransesterification+activities+were+1205.67+U%2FmL+and+1108.80+U%2FmL%2C+respectively.%0D%0AThe+immobilization+of+Klebsiella+sp.+LPG172+lipase+using+natural+zeolite%0D%0Afrom+Lampung+altered+the+optimal+conditions+for+both+enzyme+activities.+For+the%0D%0Ahydrolysis+reaction%2C+free+lipase+had+optimum+conditions+at+pH+7%2C+a+temperature+of%0D%0A80%C2%B0C%2C+and+an+incubation+time+of+25+minutes.+However%2C+after+immobilization%2C+the%0D%0Alipase+worked+optimally+at+pH+8%2C+a+temperature+of+50%C2%B0C%2C+and+an+incubation+time+of%0D%0A15+minutes.+In+the+transesterification+reaction%2C+free+lipase+had+optimum+conditions%0D%0Aat+pH+7%2C+a+temperature+of+70%C2%B0C%2C+and+an+incubation+time+of+25+minutes.+After%0D%0Aimmobilization%2C+the+lipase+functioned+optimally+at+pH+8%2C+a+temperature+of+60%C2%B0C%2C+and%0D%0Aan+incubation+time+of+10+minutes.%0D%0AThe+optimal+conditions+obtained+were+used+to+test+the+stability+of+the%0D%0Aimmobilized+lipase+against+repeated+use.+In+the+hydrolysis+reaction%2C+the+immobilized%0D%0Alipase+could+be+used+for+2+cycles+with+the+final+activity+reducing+to+43.09%25+of+its%0D%0Ainitial+activity.+In+the+transesterification+reaction%2C+the+immobilized+lipase+could+be%0D%0Aused+for+3+cycles+with+a+final+activity+percentage+of+55.98%25.%0D%0A%0D%0AKeywords%3A+hydrolysis%2C+i
