Latar Belakang: Pengobatan kanker terus berkembang pesat seiring dengan
masifnya kejadian kanker di dunia. Salah satu potensi yang dikembangkan saat ini
untuk pengobatan dan pencegahan kanker adalah antioksidan alami. Beberapa
penelitian terdahulu melaporkan bahwa adanya aktivitas antioksidan yang tinggi
pada tanaman kabau (Archidendron bubalinum). Namun, belum ada penelitian yang
menghubungkan tanaman kabau dengan potensinya sebagai antikanker, seperti
kanker payudara yang prevalensinya tinggi di dunia. Sehingga, dalam penelitian ini
dilakukan eksplorasi aktivitas antioksidan dan aktivitas sitotoksik pada sel kanker
payudara MCF-7. 
Metode: Dalam penelitian ini, proses ekstraksi dilakukan dengan metode
Ultrasound-Assisted Extraction menggunakan pelarut etanol 96%. Ekstrak kental
selanjutnya dilakukan identifikasi senyawa secara kualitatif dan diteliti aktivitas
antioksidannya menggunakan metode DPPH. Kemudian, dilakukan uji aktivitas
sitotoksik pada sel kanker payudara MCF-7 menggunakan metode MTT. 
Hasil: Ekstrak etanol kulit biji kabau mengandung senyawa saponin, fenolik, tanin,
dan flavonoid. Didapatkan hasil nilai IC
50 
aktivitas antioksidan yang tinggi sebesar
38,23 mg/L. Tidak didapatkan adanya aktivitas sitotoksik yang berarti bahwa
sampel tidak mencapai penghambatan 50% pada sel kanker payudara MCF-7. 
Simpulan: Ekstrak etanol kulit biji kabau memiliki aktivitas antioksidan yang
sangat baik, tetapi tidak menunjukkan aktivitas sitotoksik pada sel kanker payudara
MCF-7.

Background: Cancer treatment continues to evolve rapidly in line with the
increasing incidence of cancer worldwide. One potential approach currently being
developed for cancer prevention and treatment is the use of natural antioxidants.
Previous studies have reported high antioxidant activity in the Archidendron
bubalinum. However, no studies have linked this plant to its anticancer potential,
particularly against breast cancer, which has a high prevalence globally. Therefore,
this study aimed to explore the antioxidant and cytotoxic activities of Archidendron
bubalinum seed shells extract on MCF-7 breast cancer cells.
Methods: In this study, extraction was performed using the Ultrasound-Assisted
Extraction method with 96% ethanol as the solvent. The crude extract was then
qualitatively analyzed for its compound content and its antioxidant activity was
evaluated using the DPPH method, the cytotoxic activity against MCF-7 breast
cancer cells was assessed using the MTT assay.
Result: The ethanolic extract of kabau seed shells contained saponins, phenolics,
tannins, and flavonoids. The results showed a high IC
50
 value for antioxidant
activity at 38,23 mg/L. No significant cytotoxic activity was observed, indicating
that the sample did not achieve 50% inhibition of MCF-7 breast cancer cells.
Conclusion: The ethanolic extract of kabau seed shells exhibited excellent
antioxidant activity, but did not demonstrate cytotoxic activity against MCF-7
breast cancer cells.
