"Drug-resistant bacteria pose a serious risk to people's health, making it necessary to\r\nsearch for new antibacterials from natural sources such as marine sponges. The\r\nbioactive compounds of sponges are mostly produced by microbial symbionts,\r\nespecially actinobacteria, which are well-known for their high G+C content and\r\nvariety of bioactive compounds. Pseudonocardia carboxydivoran is one species of\r\nmarine actinobacteria that has the potential to produce bioactive compounds. These\r\ncompounds are often complex polyketides synthesised by the polyketide synthase\r\n(PKS) gene. Therefore, screening of PKS genes can reveal the potential for new\r\nbioactive compounds. This study aimed to isolate the polyketide synthase (PKS)\r\ngene sequence from the marine actinobacteria Pseudonocardia carboxydivorans\r\n18A13O1 associated with sponges. Samples were enriched on 1% colloidal chitin\r\nagar medium, followed by microscopic observation to ensure the purity of the\r\nisolates. Afterwards, the isolates were subcultured on liquid ISP-2 medium and\r\nincubated for 7 days. DNA was extracted using PROMEGA Kit and analysed for\r\nconcentration and purity with a nanophotometer. Subsequently, PKS sequences\r\nwere amplified using primers PKS1. Amplification results were visualised by 1.2%\r\nagarose gel electrophoresis. Sequencing was performed by the Sanger method.\r\nSequencing results were analysed using BioEdit and Open Reading Frame (ORF)-\r\nFinder from NCBI. The results showed that the PKS gene sequences obtained\r\nencoded hexokinase proteins, which are known to contribute to the production of\r\nantibiotic compounds.\r\n\r\nKeywords: Pseudonocardia carboxydivorans, antibacterial, PKS, PCR,\r\n\r\nHexokinase\r\n\r\nBakteri resisten obat menimbulkan risiko serius bagi kesehatan masyarakat,\r\nsehingga diperlukan pencarian antibakteri baru dari sumber alami seperti spons laut.\r\nSenyawa bioaktif spons sebagian besar dihasilkan oleh mikroba simbion,\r\nkhususnya aktinobakteri, yang terkenal dengan kandungan G+C tinggi dan variasi\r\nsenyawa bioaktifnya. Pseudonocardia carboxydivoran merupakan salah satu jenis\r\naktinobakteri laut yang berpotensi menghasilnya senyawa bioaktif. Senyawa ini\r\nsering merupakan poliketida kompleks yang disintesis oleh gen polyketide synthase\r\n(PKS). Oleh karena itu, skrining gen PKS dapat mengungkap potensi senyawa\r\nbioaktif baru. Penelitian ini bertujuan mengisolasi sekuen gen polyketide synthase\r\n(PKS) dari aktinobakteri laut Pseudonocardia carboxydivorans 18A13O1 yang\r\nbersimbiosis dengan spons. Sampel diremajakan pada media 1% koloid kitin agar,\r\ndilanjutkan dengan pengamatan mikroskopis untuk memastikan kemurnian isolat.\r\nKemudian, isolat disubkultur pada media ISP-2 cair dan diinkubasi selama 7 hari.\r\nDNA diekstraksi menggunakan Kit PROMEGA dan dianalisis konsentrasi dan\r\nkemurniannya dengan nanofotometer. Kemudian sekuen PKS diamplifikasi\r\nmenggunakan primer PKS1. Hasil amplifikasi divisualisasi dengan elektroforesis\r\ngel Agarosa 1,2%. Sekuensing dilakukan dengan metode Sanger. Hasil sekunsing\r\ndianalisis menggunakan BioEdit dan laman bioinformatika Open Reading Frame\r\n(ORF)-Finder NCBI.
