Lobster hemolymph clots quickly, requiring an alternative anticoagulant like fucoidan. This study aimed to evaluate the anticoagulant activity of fucoidan from brown algae using the Lee-White coagulation test, agglutination test, and gene expression analysis in lobster hemolymph for detecting endotoxins from bacteria that cause diseases in fish. This study employs an exploratory method by screening different types and doses of anticoagulants using six treatments: control, addition of 10\% aspirin, 10\% EDTA, 10\% sodium citrate, heparin, and fucoidan to lobster hemolymph. The coagulation time of the lobster hemolymph was measured, and an agglutination test was observed after the addition of fish pathogenic bacteria. The results of the Lee-White test showed that the hemolymph-to-fucoidan ratios of 2:3, 2:4, and 2:5 influenced coagulation. The agglutination test results indicated agglutination formation in all samples. The cycle threshold (CT) values for gene expression analysis were as follows: {\ensuremath{\beta}}-Actin 1 at 34.9226, {\ensuremath{\beta}}-Actin 2 at 29.8296, GPO at 42.042, Lectin 1 at 41.937, Lectin 2 at 42.1846, LGBP 1 at 39.9524, and LGBP 2 at 35.8335. In conclusion, fucoidan can be used as an anticoagulant at a 2:3 ratio, and lobster hemolymph has the potential to be utilized as an antibacterial agent against fish pathogens. Additionally, the genes {\ensuremath{\beta}}-Actin, LGBP, Lectin, and GPO were successfully expressed in the hemolymph of the sand lobster (Panulirus homarus).
