description:    Background: Takokak fruit contains antioxidant compounds with antidiabetic  properties. This study aims to determine the effect of administering ethanol extract  of takokak fruit and increasing its dosage on the area of the Langerhans islets in  the pancreas of male white rats with a diabetes mellitus model.  Methods: This experimental study used a Post Test Only Control Group Design  with a randomized controlled trial. A total of 30 male white rats were divided into  5 groups: the KN group (no treatment), the K(-) group (induced with 140 mg/Kg  body weight of alloxan), and the P1, P2, and P3 groups (induced with alloxan 140  mg/Kg body weight, then treated with ethanol extract of takokak fruit at doses of  100 mg/kgBW, 200 mg/kgBW, and 400 mg/kgBW) for 28 days. Preparations were  observed using a light microscope at 400x magnification in 5 fields of view, then  measured using the ImageJ application.  Results: The average area of the islets of Langerhans in the pancreas of male  white rats was as follows: KN group: 0.018979 mm², K(-) group: 0.002328 mm²,  P1 group: 0.004873 mm², P2 group: 0.007529 mm², and P3 group: 0.008585 mm².  Significant differences in the average area were found between the KN group and  the K(-) and P1 groups; between the K(-) group and the P1, P2, and P3 groups;  and between the P1 and P3 groups.  Conclusion: There is an effect of ethanol extract of takokak fruit on the area of  the islets of Langerhans in the pancreas of male white rats with a diabetes mellitus  model. There is an effect of increased doses of ethanol extract of takokak fruit on  the area of the islets of Langerhans in the pancreas of male white rats with a  diabetes mellitus model, with the most significant dose being 400 mg/kg body  weight.    Keywords: alloxan, diabetes mellitus, pancreas, Solanum torvum   Latar Belakang: Buah takokak mengandung senyawa antioksidan sebagai anti  diabetes. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak  etanol buah takokak dan peningkatan dosisnya terhadap luas pulau Langerhans  pankreas tikus putih jantan model diabetes melitus.  Metode: Penelitian eksperimental ini menggunakan metode rancangan acak  terkontrol Post Test Only Control Group Design. Sebanyak 30 ekor tikus putih  jantan dibagi menjadi 5 kelompok: kelompok KN sebagai tikus tanpa perlakuan;  kelompok K(-) yang diinduksi aloksan 140 mg/KgBB; kelompok P1, P2, dan P3  yang diinduksi aloksan 140 mg/KgBB, kemudian diberi ekstrak etanol buah  takokak dengan dosis berbeda (100 mg/kgBB, 200 mg/kgBB, dan 400 mg/kgBB)  selama 28 hari. Pengamatan preparat menggunakan mikroskop cahaya dengan  perbesaran 400x dalam 5 lapang pandang kemudian diukur menggunakan aplikasi  ImageJ.  Hasil: Didapatkan rerata luas pulau Langerhans pankreas tikus putih jantan   kelompok KN: 0,018979 mm  2  , K(-): 0,002328 mm  2  , P1: 0,004873 mm  2  , P2:  0,007529 mm  2  , dan P3: 0,008585 mm  2  . Didapatkan perbedaan rerata yang  signifikan antara kelompok KN dengan K(-) dan P1; kelompok K(-) dengan P1,  P2, dan P3; dan P1 dengan P3.  Simpulan: Terdapat pengaruh dari pemberian ekstrak etanol buah takokak  terhadap luas pulau Langerhans pankreas tikus putih jantan model diabetes  melitus. Terdapat pengaruh dari peningkatan dosis ekstrak etanol buah takokak  terhadap luas pulau Langerhans pankreas tikus putih jantan model diabetes  melitus, dengan dosis yang paling berpengaruh adalah 400 mg/kgBB.
