ASPERGILLUS ACULEATUS ISOLAT TANAH HUMUSAbstrak
Tujuan penelitian ini adalah untuk mengisolasi dan memurnikan enzim
kitin deasetilase dari Aspergillus aculeatus isolat tanah humus. Kitin deasetilase
dari Aspergillus aculeatus isolat tanah humus menggunakan kitin sebagai sumber
karbon yang ada di dalam medium fermentasi padat dan dishaker selama 5 hari
pada suhu ruang . Pemurnian kitin deasetilase meliputi pengendapan amonium
sulfat dan dialisis.
Fraksinasi dengan amonium sulfat mempunyai nilai kadar proteinnya
sebesar 3,47 mg/ml serta pemurnian tahap selanjutnya yaitu dialisis yang
mempunyai kadar protein sebesar 2,93 mg/ml. Hasil analisis spectra IR
menunjukkan terjadinya perubahan peak di spektra IR glikol kitin yaitu adanya
gugus fungsi amida dan di spektra deasetilase substrat glikol kitin dengan enzim
kitin deasetilase adanya gugus fungsi amina primer.

Abstract
The aims of the research were to isolate and purify chitin deacetylase
which produced by Aspergillus aculeatus from humus soil. Chitin deacetylase
from Aspergillus aculeatus has been produced using chitin as sole carbon resource
of solid fermentation medium shaked for 5 day in room temperature. The purified
chitin deacetylase through ammonium sulfate precipitation and dialysis.
The result, ammonium sulfate precipitation showed that the highest lowry
activity of chitin deacetylase was 3,47 mg/ml in fraction 40-60% and dialysis
showed that the lowry activity of chitin deacetylase was 2,93 mg/ml. Analysed of
IR peak of glycol chitin was amide and in peak deacetylation of glycol chitin with
chitin deacetylation enzyme was primary amine.
