RESISTEN TERHADAP CEKAMAN GARAM (NaCl)
SECARA IN VITROPisang raja bulu ditemukan di Desa Cibereum, Kecamatan Cisarua, Provinsi
Jawa Barat. Pisang raja bulu merupakan jenis pisang yang memiliki nilai
ekonomis tinggi untuk dikembangkan di Indonesia sehingga perlu dilakukan
pengujian tanaman pisang raja bulu yang toleran atau resisten terhadap
cekaman lingkungan salah satunya yaitu cekaman garam. Cekaman adalah
keadaan dimana terjadi perbedaan keseimbangan antara unsur biotik dan
abiotik. Cekaman garam pada tanaman bisa mengakibatkan pertumbuhan
tidak normal, daun kecil dan terbakar, pertumbuhan kerdil, buah tidak
sempurna, dan hasil menurun. Tujuan penelitian ini adalah mengetahui
konsentrasi NaCl yang efektif terhadap pertumbuhan planlet pisang raja bulu
yang resisten terhadap cekaman garam secara in vitro. Penelitian ini
dilaksanakan di ruang Kultur In Vitro, Laboratorium Botani, Jurusan Biologi,
Fakultas Matematika dan Ilmu Pengetahuan Alam, Universitas Lampung,
menggunakan medium MS padat dan penambahan NaCl. Penyusunan
rancangan penelitian secara faktorial menggunakan pola dasar Rancangan
Acak Lengkap (RAL) yang terdiri dari faktor tunggal yaitu konsentrasi NaCl
dengan 5 taraf perlakuan : P0 (0%), P1(0,25%), P2 (0,50%), P3 (0,75%), dan
P4 (1%). Masing-masing perlakuan dilakukan 5 kali ulangan. Parameter yang
diamati yakni jumlah daun, jumlah tunas, tinggi planlet, berat basah, dan
visualisasi planlet serta kandungan klorofil. Homogenitas ragam di uji
menggunakan uji Levene dan dilanjutkan dengan uji Anova one way pada
taraf nyata 5%. Hasil dari penelitian yang telah dilakukan menunjukkan
bahwa konsentrasi NaCl yang efektif untuk pertumbuhan pisang raja bulu
yang resisten terhadap cekaman garam yaitu 0,50%

food developed in Indonesia, so it is necessary to test the banana king Bulu plant

between biotic and abiotic elements. Salt stress on plants can result in abnormal
growth, small leaves and burning, stunted growth, incomplete fruit, and decreased
yields. The purpose of this research is to determine the effective concentration of
NaCl on the growth of banana plantlets king of the feathers that are resistant to
salt stress in vitro. This research carried out in the In Vitro Culture room,
Botanical Laboratory, Department of Biology, Faculty of Mathematics and
Natural Sciences, University of Lampung, using the medium Solid MS and
addition of NaCl. Factorial research design preparation using the basic pattern of
Completely Randomized Design (CRD) consisting of single, namely the
concentration of NaCl with 5 levels of treatment:,P1(0.25%), P2 (0.50%), P3
(0.75%), and P4 (1%). Each treatment was repeated 5 times. Parameters observed
were number of leaves, number of shoots, plantlet height, wet weight, and
visualization of plantlets and chlorophyll content. Homogeneity of variance was
tested using the test Levene and continued with the one-way Anova test at a 5%
significance level. Result of research that has been done shows that Based on the
results of research and From the discussion it can be concluded that the effective
concentration of NaCl for selection of banana plantains that are resistant to salt
stress, namely a concentration of 0.50%
