STRAIN I18 DAN SERRATIA MARCESCENS STRAIN MBC1 SEBAGAI
OVISIDA NYAMUK AEDES AEGYPTIPenyakit Demam Berdarah Dengue (DBD) merupakan salah satu penyakit
endemik dan masih menjadi permasalahan kesehatan di Indonesia. Virus Dengue
ini ditularkan melalui gigitan nyamuk, yaitu Aedes aegypti, Aedes albopictus dan
Aedes scutellaris. Vektor utama atau nyamuk yang lebih berperan dalam
menularkan virus Dengue adalah nyamuk Ae. aegypti. Untuk mengatasi
permasalah tersebut dilakukan pengendalian vektor secara hayati dengan
memanfaatkan potensi metabolit sekunder yang dihasilkan oleh mikroorganisme.
Ekstrak metabolit ini digunakan sebagai ovisida telur nyamuk Ae. aegypti untuk
menurunkan kepadatan nyamuk. Tujuan penelitian ini adalah untuk melakukan
uji potensi dan mengetahui kefektifan (nilai LC50 dan LC90) metabolit sekunder
dari bakteri Serratia marcescens strain MBC1 dan Streptomyces sp. strain I18
koleksi Laboratorium Mikrobiologi Fakultas Matematika dan Ilmu Pengetahuan
Alam dengan empat konsentrasi, yaitu 125 ppm, 250 ppm, 500 ppm dan 1.000
ppm sebagai ovisida terhadap telur nyamuk Ae. aegypti. Metode yang
digunakan yaitu dengan rancangan desain faktorial 2x4 dan digunakan 25 telur
nyamuk Ae. aegypti dengan 4 kali pengulangan menggunakan air sumur sebagai
kontrol negatif dan aseton sebagai kontrol positif. Hasil penelitian ini
menunjukkan bahwa ekstrak metabolit sekunder dari Serratia marcescens strain
MBC1 mampu menghambat penetasan telur nyamuk Ae. aegypti. Ekstrak
metabolit sekunder dari Serratia marcescens strain MBC1 dan Streptomyces sp.
strain I18 mampu menghambat siklus hidup Ae. aegypti ditandai dengan adanya
kematian larva instar III pada konsentrasi 1.000 ppm.
Potensi yang dimiliki oleh Serratia marcescens strain MBC1 dapat dijadikan
sebagai kandidat ovisida nyamuk Ae. aegypti.

Dengue Hemorrhagic Fever (DHF) is one of the endemic diseases and is still a
health problem in Indonesia. Dengue virus is transmitted through mosquito bites,
namely Aedes aegypti, Aedes albopictus and Aedes scutellaris. The main vector or
mosquito that plays a role in transmitting the dengue virus is Ae. aegypti. To
overcome this problem, biological vector control is carried out by utilizing the
potential of secondary metabolites produced by microorganisms. This metabolite
extract was used as an oviside for the eggs of Ae. aegypti to reduce mosquito
density. The purpose of this study was to test the potency and determine the
effectiveness (LC50 and LC90 values) of secondary metabolites of the bacteria
Serratia marcescens strain MBC1 and Streptomyces sp. strain I18 collection of
the Laboratory of Microbiology, Faculty of Mathematics and Natural Sciences
with four concentrations, namely 125 ppm, 250 ppm, 500 ppm and 1,000 ppm as
ovicide against Ae. aegypti. The method used is a 2x4 factorial design and 25 eggs
of Ae. aegypti with 4 repetitions using water well as a negative control and
acetone as a positive control. The results of this study showed that the secondary
metabolite extract of Serratia marcescens strain MBC1 was able to inhibit the
hatching of Ae. aegypti. Extracts of secondary metabolites from Serratia
marcescens strain MBC1 and Streptomyces sp. strain I18 was able to inhibit the
life cycle of Ae. aegypti was characterized by the death of third instar larvae at a
concentration of 1,000 ppm. The potential possessed by Serratia marcescens
strain MBC1 can be used as a ovicides of Ae. aegypti.
