Abstrak

Senyawa karsinogenik dapat memicu terbentuknya kanker (karsinogenesis) dengan fase awal berupa pembentukan lesi pada DNA, perusakan jaringan, perubahan-perubahan sistem imun, susunan protein tubuh dan biokomiawi sel tubuh. Tumbuhan Jeruju (A. ilicifolius), Lamun (E. acoroides) dan asam organik Taurin merupakan senyawa alami dengan potensi antioksidan dan antikanker sebagai agen kemopreventif yang dapat menghambat proses karsinogenesis. Tujuan dari penelitian ini adalah menguji efek ekstrak metanol daun jeruju (A. ilicifolius) dan lamun (E. acoroides), serta taurin terhadap profil protein plasma darah, dan histopatologi organ hepar mencit jantan (Mus musculus L.) yang diinduksi karsinogenik benzo(α)piren. Penelitian ini dilakukan dengan Rancangan Acak Lengkap (RAL) menggunakan 25 ekor mencit jantan yang dibagi dalam 5 kelompok dengan masing-masing 5 ulangan. Pada K2, K3, K4 dan K5 diinduksi benzo(α)piren secara subkutan dengan dosis 0,3 mg/bb selama 10 hari. Kemudian selama 15 hari setelahnya, K3 diberikan ekstrak daun jeruju dosis 17,5 mg/bb/hari, K4 diberikan ekstrak lamun dosis 8,7 mg/bb/hari, dan K5 diberikan taurin sebanyak 15,6mg/bb/hari. Hasil penelitian menunjukkan pemberian keseluruhan bahan uji (ekstrak metanol jeruju, ekstrak metanol lamun dan taurin) belum dapat menunjukan pengaruh terhadap profil protein plasma darah mencit yang diinduksi benzo(α)piren, namun dapat menurunkan tingkat kerusakan pada histopatologi hepar mencit yang diinduksi benzo(α)piren. Hasil pengukuran parameter lainnya menunjukkan bahwa pemberian keseluruhan bahan uji mampu menurunkan rerata jumlah total leukosit dan persentase limfosit mencit, serta meningkatkan persentase neutrofil mencit, namun belum dapat menunjukan pengaruh terhadap berat badan mencit, rerata jumlah total eritrosit mencit, rerata kadar protein total plasma darah mencit, rerata berat organ dan rerata indeks organ hepar mencit yang diinduksi benzo(α)piren. Kata kunci : jeruju, lamun, taurin, profil protein, histopatologi hepar, benzo(α)piren ABSTRACT Carcinogenic compounds can cause cancer (carcinogenesis) with the initial phase such as the formation of lesions in DNA, tissue destruction, disruption of the immune system, protein structure and cell regulation. Jeruju (A. ilicifolius), Seagrass (E. acoroides) and Taurine (organic acid) are natural compounds with antioxidant and anticancer potential as chemopreventive agents that can inhibit the carcinogenesis. The purpose of this study was to examine the effect of methanolic extract of jeruju leaves (A. ilicifolius) seagrass (E. acoroides), and taurine on the profile of blood plasma proteins, and histopathology of liver in male mice (Mus musculus L.) induced by benzo(α)pyrene as a carcinogenic. This study was carried out in a completely randomized design (CRD) using 25 male mice which were divided into 5 groups with 5 replications on each. K2, K3, K4 and K5, was induced by benzo(α)pyrene subcutaneously 0.3 mg/bw for 10 days. Then 15 days thereafter, K3 was given the jeruju leaves methanolic extract 17.5 mg/bw/day, K4 was given the seagrass leaves methanolic extract 8.7 mg/bw/day, and K5 was given taurine 15.6 mg/bw/day. The results showed that the whole test material (methanolic extract of jeruju leaves, methanolic extract of seagrass and taurine) had not been able to show influence on the profile of blood plasma proteins, but could reduce the level of liver damage of male mice induced by benzo(α)pyrene. The results of other parameters showed that the overall administration of the test material was able to reduce the mean total number of leukocyte and the percentage of lymphocytes, and increase the percentage of neutrophils, but could not show the effect of body weight, the mean total number of erythrocytes, the mean of blood plasma concentration, the mean of liver weight and the mean of liver index of male mice induced by benzo(α)pyrene.
