Abstrak

This study was aimed to reveal the identity of Aspergillus spp. and Talaromyces spp. and their ability as antagonist, phosphate solubilizing fungi, chitin degrading fungi, Plant Growth Promoting Fungi (PGPF) and their aflatoxin production. Four isolates of Aspergillus spp. and six isolates of Talaromyces spp. were used in this study. Those all isolates were obtained from three different plants rhizosphere, namely pineapple (AS1, AS2, AS3, AS4, AS5), corn (AS6, AS7, AS8, AS9), and chili (AS11). Identification (morphological characteristics and molecular technique), antagonist test against Phytophthora palmivora, investigation on ability as phosphate solubilizing fungi or chitin degrading fungi as well as aflatoxin production were conducted at Agricultural Biotecnology, Faculty of Agriculture, University of Lampung. The ability as PGPF was tested using cucumber plant as indicator plant at green house of Faculty of Agriculture. The data collected in this study were identity of the isolates, percentage of inhibition against P. palmivora, clear (halo) zone area around the colony which was grown on pikovskaya and chitin agar media. As for their ability as Plant Growth Promoting Fungi (PGPF), some parameters were observed namely plant height, wet weight of shoot and root, dry weight of shoot and root, greenish leaves and percentage of shoot N and P content and fluorescence of the colony grown on CAM media visualized under UV. Based on the morphological characteristics, it was confirmed that all the isolates were in the group of Aspergillus and Talaromyces. Sequence analysis result of ITS region (ITS1-ITS4) revealed that 4 isolates (AS1, AS6, AS7, and AS9) were placed in the group of Aspergillus oryzae and 6 isolates (AS2, AS3, AS4, AS5, AS8, and AS11) were palced in to group of Talaromyces sayulitensis. Isolate AS1, AS4, AS5, AS8, and AS11 has capability as antagonist of P. palmivora, phosphate solubilizing fungi, chitin degrading fungi, and PGPF. All the isolates doesn’t have ability to product aflatoxin. Keywords: Antagonist, Aspergillus oryzae, Plant Growth Promoting Fungi, Talaromyces sayulitensis. ABSTRAK STUDI KOMPREHENSIF JAMUR Aspergillus spp. DAN Talaromyces spp. Penelitian ini bertujuan mengidentifikasi Aspergillus spp. dan Talaromyces spp. dan kemampuannya sebagai antagonis, pelarut fosfat, pendegradasi kitin, Plant Growth Promoting Fungi (PGPF), dan produksi aflatoksin. Empat isolat Aspergillus spp. dan enam isolat Talaromyces spp. diisolasi dari rizosfer tanaman nanas (AS1, AS2, AS3, AS4, AS5), tanaman jagung (AS6, AS7, AS8, AS9), dan tanaman cabai (AS11). Identifikasi (karakter morfologi dan teknik molecular), uji antagonis terhadap Phytophthora palmivora, kemampuan jamur sebagai pelarut fosfat dan pendegradasi kitin, serta produksi aflatoksin jamur dilakukan di Laboratorium Bioteknologi Fakultas Pertanian (LBFP), Universitas Lampung. Kemampuan jamur diuji sebagai PGPF menggunakan tanaman mentimun sebagai tanaman indikator dilakukan di rumah kaca Fakultas Pertanian. Data yang diamati meliputi identitas semua isolat, persentasi penghambatan P. palmivora, zona bening yang tumbuh di media pikovskaya dan kitin agar. Serta kemampuan jamur sebagai Plant Growth Promoting Fungi (PGPF), parameter yang diamati diantaranya tinggi tanaman, bobot basah tajuk dan akar, bobot kering tajuk dan akar, kehijauan daun dan penyerapan N dan P pada tajuk dan sinar fluorescence pada koloni yang ditumbuhkan di media CAM divisualisasi di bawah sinar UV. Berdasarkan karakteristik morfologinya, dikonfirmasi bahwa semua isolat masuk dalam jamur Aspergillus dan Talaromyces. Analisis sekuensing menggunakan ITS region menunjukkan bahwa 4 isolat (AS1, AS6, AS7, dan AS9) masuk ke dalam spesies Aspergillus oryzae dan 6 isolat (AS2, AS3, AS4, AS5, AS8, dan AS11) masuk ke dalam spesies Talaromyces sayulitensis. Isolat AS1, AS4, AS5, AS8, dan AS11 memiliki kemampuan menghambat pertumbuhan P. palmivora, pelarut fosfat, pendegradasi kitin, serta sebagai PGPF. Semua isolat tidak memiliki kemampuan untuk memproduksi aflatoksin. Kata Kunci: Antagonis, Aspergillus oryzae, Plant Growth Promoting Fungi, Talaromyces sayulitensis.
