Abstrak

Penelitian ini dilakukan untuk melihat jumlah produk glukosamin dari kerja enzim kitinase dan kitin deasetilase yang dihasilkan Mucor miehei. Degradasi kitin menjadi glukosamin oleh 2 enzim tersebut berlangsung melalui reaksi fermentasi. Proses fermentasi diamati 1, 2, 3, 4, 8, 12 dan 24 jam. Jumlah produk glukosamin hasil fermentasi tertinggi dari fraksi freeze-dry didapatkan pada waktu inkubasi 2 jam sebesar 82,2%. Kadar glukosamin dari hasil fermentasi ditentukan dengan metode spektrofotometri UV-Vis. Hasil analisis secara spektrofotometri UV-Vis menunjukkan bahwa kadar glukosamin tertinggi didapatkan pada waktu inkubasi 3 jam sebesar 0,01667%. Kata kunci : Mucor miehei, spektrofotometer UV-Vis, Kitin, Glukosamin, Kitinase, Kitin Deasetilase ABSTRACT This research was conducted to measure the amount of glucosamine product on the working of chitinase and chitin deacetylase enzyme which is produced by Mucor miehei. Degradation from chitin to glucosamine by 2 enzymes occur through fermentation reaction. Fermentation process was observed at 1, 2, 3, 4, 8, 12 and 24 hours. The highest amount of glucosamine product as freeze-dry fraction was obtained for 82,2% at 2 hours incubation time. The glucosamine level from fermentation yield was determined with spectrophotometry UV-Vis method. The results of the analysis in spectrophotometry UV-Vis were showed that the highest glucosamine level was obtained for 0,01667% at 3 hours incubation time.
