Abstract
ABSTRAK Penelitian ini bertujuan untuk mengetahui pengaruh jarak straw dengan nitrogen cair pada proses pre freezing serta jarak straw yang terbaik terhadap kualitas semen beku Sapi Limousin sehingga layak untuk proses IB. Penelitian ini dilaksanakan di Unit Pelayanan Teknis Daerah Balai Insemninasi Buatan Daerah (UPTD-BIBD) Lampung, Kecamatan Terbanggi Besar, Kabupaten Lampung Tengah, Provinsi Lampung pada tanggal 07—13 April 2014. Rancangan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan 5 perlakuan jarak straw, yaitu 2 cm, 4 cm, 6 cm, 8 cm, dan 10 cm dengan 4 kali ulangan. Data yang diperoleh dianalisis menggunakan analisis ragam dan dilanjutkan dengan uji Polinomial Ortogonal. Hasil penelitian menunjukkan bahwa jarak straw berpengaruh sangat nyata (P<0,01) terhadap motilitas dan spermatozoa hidup setelah thawing namun tidak berpengaruh nyata (P>0,05) terhadap motilitas dan spermatozoa hidup setelah pre freezing. Uji polinomial ortogonal diperoleh bahwa jarak straw berpengaruh sangat nyata (P<0,01) terhadap motilitas spermatozoa setelah thawing, berpola kuadratik dengan persamaan Y = - 6,50 + 12,09x – 0,89x2 ; R2 = 41,3%, dengan pengaruh yang terbaik pada jarak straw 6 cm yaitu 34%. Selain itu jarak straw juga berpengaruh sangat nyata (P<0,01) terhadap persentase spermatozoa hidup setelah thawing, berpola kuadratik dengan persamaan Y = 8,97 + 8,81x - 0,64x2 ; R2 = 41,3%, dengan pengaruh yang terbaik pada jarak straw 6 cm yaitu 38,79%. Kata kunci: kualitas semen, Sapi Limousin, jarak straw ABSTRACT This study aimed to determine the effect of straw space with liquid nitrogen on pre freezing process and the best straw space to the quality of frozen semen of Limousin Cows so that they are appropriate for Artificial Insemination process. This study was conducted in Regional Technical Service Unit-Regional Artificial Insemination Office Lampung, Terbanggi Besar District, Central Lampung Regency, Lampung Province on 7th—13 th April 2014. The Completely Randomized Design (CRD) was used is these research with five treatments of straw spaces, 2 cm, 4 cm, 6 cm, 8 cm, and 10 cm, with 4 replications. The data obtained was analyzed by using variance analysis andcontinued by Polinomial Ortogonal test. The result of this study showed that the straw space was significantly (P<0,01) to the motility and live spermatozoa after thawing. However it was not significantly (P>0,05) to the motility and live spermatozoa after pre freezing. Polinomial Ortogonal test, obtained that the straw space was significantly (P<0,01) to the motility and live spermatozoa after thawing, had quadratic pattern with equation of Y = - 6,50 + 12,09x – 0,89x2 ; R2 = 41,3%, with the best effect on the straw space of 6 cm, that is 34%. Besides, the straw space was also different (P<0,01) to percentage of live spermatozoa after thawing, had quadratic pattern with equation of Y = 8,97 + 8,81x - 0,64x2 ; R2 = 41,3%, with the best effect on the straw space of 6 cm, that is 38,79%.
