Abstrak

Lobster air tawar (Cherax quadricarinatus) merupakan komoditas budidaya air tawar yang saat ini diminati masyarakat karena memiliki morfologi yang menarik dan nilai kandungan gizi yang tinggi sehingga perlu dibudidayakan. Untuk meningkatkan produksinya dapat dilakukan dengan budidaya monoseks jantan dengan metode sex reversal. Penelitian ini bertujuan untuk: 1)mengetahui dosis dan lama perendaman yang efektif juvenil lobster air tawar (Cherax quadricarinatus) yang diberi ekstrak steroid teripang pasir (Holothuria scabra Jaeger) terhadap kadar testosteron dalam hemolymph 2)mengetahui interaksi antara dosis dan lama perendaman pada ekstrak steroid teripang pasir (Holothuria scabra Jaeger) pada juvenil lobster air tawar terhadap kadar testosteron dalam hemolymph. Penelitian ini menggunakan Rancangan Acak Lengkap Faktorial dua variabel yang terdiri dari dosis 0, 2, dan 4 ppm sedangkan lama perendaman 18 dan 30 jam. Perlakuan sebanyak 8 kelompok dan masing-masing diulang sebanyak 3 kali. Data dianalisis dengan Sidik Ragam dan jika terdapat perbedaan nyata maka dilanjutkan dengan uji Beda Nyata Terkecil (BNT) dengan taraf 5%. Hasil penelitian menunjukkan variasi dosis dan lama perendaman berpengaruh terhadap peningkatan kadar testosteron. Perendaman dalam ekstrak steroid teripang pasir memberikan pengaruh nyata terhadap meningkatnya kadar testosteron dan tingkat keberhasilan pembentukan kelamin jantan lobster air tawar. Rerata kadar testosteron tertinggi sebesar 0,248 ng/mL dengan persentase pembentukan jantan tertinggi sebesar 77,45% terdapat pada perlakuan pemberian ekstrak steroid teripang pasir dosis 4 ppm dan lama perendaman 30 jam. Kata kunci: testosteron, lobster air tawar, sex reversal, steroid, teripang pasir ABSTRACT Crayfish (Cherax quadricarinatus) is a freshwater aquaculture that is currently in demand by the market, because it has an interesting morphology and high nutritional value so it need to be cultivated. To increase their production can be done by monosexual male cultivation using sex reversal method. This study aime 1) to determine the effective dose and immersion time of juvenile freshwater crayfish (Cherax quadricarinatus) which was given steroid extracts of sea cucumber (Holothuria scabra Jaeger) on the testosterone levels of the hemolymph. 2) to identify the interaction between dose and immersion time in the steroid extracts of sea cucumber (Holothuria scabra Jaeger) juvenile freshwater crayfish on their testosterone levels in the hemolymph. This study used a Completely Randomized Design Factorial (CRDF) with two variables consisting of sea cucumber of dose variations (0, 2, 4 ppm) steroid extract of sea cucumber and 2 ppm 17-α methyltestosteron with immersion time of 18 and 30 hours. There are 8 group treatments and each treatment with three replications. The data were calculated with Analyze of variance (Anova) and if there was any different then continued with the Smallest Significant Difference Test with a level of 5%. The results indicated variations in dose and immersion time gave an effect on increasing testosterone levels. Immersion in steroid extracts of sea cucumber had a significant effect on increasing testosterone levels and the success of male sex formation in the freshwater crayfish. The highest average testosterone level was 0.248 ng/mL with the highest percentage of male formation at 77.45% was found in the treatment of giving steroid extract of sea cucumber at dose of 4 ppm and 30 hours of immersion time.
