ABSTRAK UJI PATOGENISITAS DAN STUDI IN VIVO BAKTERI BIOKONTROL Bacillus sp. D2.2 TERHADAP Vibrio alginolyticus PADA PEMELIHARAAN UDANG VANAME (Litopenaeus vannamei) Oleh SERA HARDIYANI Penyakit vibriosis yang disebabkan oleh bakteri genus Vibrio, telah lama menjadi masalah utama bagi pelaku industri budidaya udang khususnya pada larva/benih udang karena dapat menyebabkan tingkat kematian larva udang melebihi 70%. Kehadiran bakteri agen biokontrol dapat dijadikan solusi bagi permasalahan pemberantasan penyakit untuk menekan pertumbuhan bakteri patogen. Penggunaan bakteri biokontrol, seperti bakteri Bacillus sp. D2.2 terbukti mampu membentuk zona hambat terhadap bakteri V. harveyi secara in vitro. Penelitian ini dilakukan untuk mengetahui tingkat patogenisitas bakteri biokontrol Bacillus sp. D2.2 terhadap udang vaname dan kemampuannya dalam menghambat pertumbuhan bakteri patogen Vibrio sp. secara in vivo. Bacillus sp. D2.2 disediakan dengan tingkat kepadatan 103, 104, 105 dan 106 CFU/ml untuk diuji patogenisitasnya melalui metode LD50. Hasil LD50 menunjukkan Bacillus sp. D2.2 tidak bersifat patogen karena tidak ada konsentrasi bakteri yang mematikan hingga 50% larva udang vaname. Selanjutnya uji antagonisme Bacillus sp. D2.2 terhadap V.alginolyticus dilakukan secara in vivo dengan 2 perlakuan yaitu pemeliharaan udang vaname dengan V.alginolyticus 105 CFU/ml dan pemeliharaan udang vaname dengan V.alginolyticus yang diuji tantang dengan Bacillus sp. D2.2 105 CFU/ml. Hasil penelitian menunjukkan Bacillus sp. D2.2 mampu menurunkan pertumbuhan V.alginolyticus dari 105 CFU/ml sampai 103 CFU/ml. hal tersebut menjelaskan bahwa Bacillus sp. D2.2 berpotensi sebagai bakteri biokontrol yang mampu menurunkan pertumbuhan bakteri patogen V.alginolyticus. Kata kunci: biokontrol, Bacillus sp. D2.2, larva udang vaname, V.alginolyticus. ABSTRACT PHATOGENICITY TEST AND IN VIVO STUDY OF BIOCONTROL BACTERIA Bacillus sp. D2.2 AGAINST Vibrio alginolyticus IN WHITE SHRIMP (Litopenaeus vannamei) REARING SYSTEM By SERA HARDIYANI Vibriosis disease caused by bacteria of the genus Vibrio, has been a major problem for the shrimp aquaculture industry, especially in the larvae / seed shrimp because it can cause larval shrimp mortality rate exceeds 70%. The presence of bacterial biocontrol agents can be used as a solution to decrease the growth of pathogenic bacteria. The use of biocontrol bacteria, such as Bacillus sp. D2.2 was proved to be able to form a zone of inhibition against V. harveyi bacteria in vitro. This study was conducted to determine the degree of pathogenicity of biocontrol bacteria Bacillus sp. D2.2 to white shrimp and its ability to inhibit the growth of pathogenic bacteria Vibrio sp. in vivo. Bacillus sp. D2.2 supplied with density 103, 104, 105 and 106 CFU/ml were tested for pathogenicity through LD50 method. The results showed that Bacillus sp. D2.2 wasn’t pathogenic because there was no bacteria concentrations that caused lethal to 50% of white shrimp larvae. Further antagonism tests of Bacillus sp. D2.2 against V.alginolyticus performed in vivo with 2 treatments, i.e white shrimps reared with additional V.alginolyticus 105 CFU/ml and white shrimps reared with additional V.alginolyticus then challenged with Bacillus sp. D2.2 at the same density. The results showed that Bacillus sp. D2.2 was able to reduce the growth of V.alginolyticus from 105 CFU/ml to 103 CFU/ml. It was explained that Bacillus sp. D2.2 has a potency as biocontrol agent bacteria to reduce pathogenic bacterial growth. Keywords: biocontrol, Bacillus sp. D2.2, white shrimp larvae, V.alginolyticus
