Background : Paracetamol is a relatively safe drug for the stomach, some studies suggest that exceeding the therapeutic dose of paracetamol can cause gastric damage. Papasan cucumber has antioxidant effects to enhance mucus and bicarbonate production. This study aims to determine the influence of ethanol extract from papasan cucumber (Coccinia grandis) on the histopathological profile of the stomach in male Sprague-Dawley rats induced by paracetamol." Method : This study follows a Post Test Only Control Group Design pattern using 25 rats divided into 5 groups with treatments administered for 10 days. The normal control group received oral aquades. The negative control group received paracetamol at a dose of 250mg/kgBW/day. The treatment group was given graded doses of ethanol extract from papasan cucumber, specifically 125, 250, and 500mg/kgBW. Result : The total gastric damage scores obtained were as follows: Normal control=11, Negative control=26, Treatment 1=23, Treatment 2=16, Treatment 3=13. The Kruskal-Wallis analysis of gastric damage followed by Post Hoc MannWhitney to examine the differences between groups revealed a significant mean difference between the negative control group and Treatment 2, as well as between the negative control group and Treatment 3 (p=0,001). Conclusion : The administration of ethanol extract from papasan cucumber at doses of 250 and 500 mg/kgBW/day for 10 days can prevent gastric damage in male white rats induced by a paracetamol dose of 250 mg/kgBW/day. The effect is more pronounced at the dose of 500 mg/kgBW/day compared to the dose of 250 mg/kgBW/day. Keywords : Gaster, Papasan Cucumber, Paracetamol Latar Belakang : Parasetamol merupakan obat yang relatif aman bagi gaster, tetapi beberapa penelitian menyatakan parasetamol yang melebihi dosis terapi dapat menyebabkan kerusakan gaster. Buah timun papasan memiliki efek antioksidan untuk meningkatkan produksi mucus dan bikarbonat. Penelitian ini bertujuan untuk mengetahui adanya pengaruh pemberian ekstrak etanol buah timun papasan (Coccinia grandis) terhadap gambaran histopatologi gaster tikus putih (Rattus norvegicus) jantan galur Sprague-Dawley yang diinduksi parasetamol. Metode : Penelitian ini merupakan pola Post Test Only Control Group Design menggunakan 25 ekor tikus yang dibagi menjadi 5 kelompok dengan perlakuan selama 10 hari. Kontrol normal diberi aquades peroral. Kontrol negatif diberi parasetamol 250mg/kgBB/hari. Perlakuan 1 diberi parasetamol 250mg/kgBB/hari dan ekstrak etanol buah timun papasan 125mg/kgBB/hari. Perlakuan 2 diberi parasetamol 250mg/kgBB/hari dan ekstrak etanol buah timun papasan 250mg/kgBB/hari. Perlakuan 3 diberi parasetamol 250mg/kgBB/hari dan ekstrak etanol buah timun papasan 500mg/kgBB/hari. Pengamatan dilakukan menggunakan mikroskop dengan melihat derajat kerusakan gaster. Hasil : Total skor kerusakan gaster yang didapat pada Kontrol normal=11, Kontrol negatif=26, Perlakuan 1=23, Perlakuan 2=16, Perlakuan 3=13. Hasil analisis Kruskal Wallis kerusakan gaster dan dilanjutkan dengan Post Hoc Mann Whitney untuk melihat perbedaan antar kelompok, diperoleh rerata yang bermakna antara kelompok kontrol negatif dengan Perlakuan 2 dan Perlakuan 3 (p=0,001). Simpulan : Pemberian ekstrak etanol buah timun papasan dosis 250 dan 500 mg/kgBB/hari selama 10 hari dapat mencegah kerusakan gaster tikus putih jantan yang diinduksi parasetamol dosis 250 mg/kgBB/hari. Pada dosis ekstrak 500 mg/kgBB/hari lebih baik efeknya dibanding dosis 250 mg/kgBB/hari. Kata kunci : Buah Timun Papasan, Gaster, Parasetamol
